Objective Src is the first discovered human oncogene, which have tyrosine kinase activity. Its clinical value has attracted a wide spread attention. In this study, we investigated the effect of the inhibitor of Src - pp2 on the growth, motility, invasion and angiogenesis of human gastric carcinoma cell line SGC - 7901. Methods Stable expression of Src and p -Src proteins was confirmed by western blot analysis Src and p - Src protein expression were tested by Immunohistochemistry; We measured effect of pp2 on proliferation p - Src protein expression of human gastric carcinoma cell line SGC - 7901 by Western blot; MTT was used to evaluate cell proliferation. Tumor invasion was examined by Transwell method, and tumor cell migration by wound - healing method. Results Immunohistochemistry revealed that Src and p - Src were expressed in the cytosol in the human gastric carcinoma cells. Western blot analysis showed that 5μmol/L pp2 had marked inhibitory effect on p - Src expression in the human gastric carcinoma cell line SGC~ 7901. When Human Gastric Carcinoma SGC~ 7901 wrere cultured with pp2 for 12 hour, the result showed that pp2 could not impact on cell proliferation at 0μmol/L(0. 33±0. 22), 5μmol/L (0. 33±0. 21) and 10 limol/L (0.33±1.02) (P>0.05), while 15 ^mol/L (3.63±1. 02) could inhibit cell prolif-eration(P<0. 05); In the membrance invasion culture system (Transwell chamber), suppressions were noted after pretreatments with pp2(5μmol/L and 10μmol/L), this effect can be enhance by the concentration gradually increased, the migration distance was (3. 92±0. 13)μm, (2. 54±0.16)μm and (1.56±0.12)μm at 0μmol/L, 5μmol/L and 10μmol/L, respectively. There were more cells invading through thematrigel in 5μmol/L and 10μmol/ L group than that in 0μmol/L group in a concentration - dependent manner [ (321. 67±28. 39) and (256.00±29.84) vs (496. 67±34.64), P< 0.05]. Angiogenesis in vitro assay showed that pp2 could inhibit the angiogenesis of human gastric carcinoma cell line at 5μmol/L and 10μmol/L compare with 0μmol/L group[ (17.33±1. 89) and (11.17±1. 49) vs( 24.5±1.71), P< 0.05 ]. Conclusions Pp2 could inhibit the growth, invasion, metastasis and angiogenesis of human gastric carcinoma cell line SGC - 7901. Src has the potential to become a new target for treatment of gastric carcinoma.
