Objective To explore the correlation between the potential mechanism of Edemaaround the hematoma and antihypertensive therapy in spontaneously hypertensiverats(SHRs).Methods A model of intracerebral hemorrhage was induced by infusion ofbacterial collagenase into the SHRs brains.The rats were randomly assigned tointracerebral hemorrhage group, amlodipine treatment group and intensive amlodipinetreatment group. The rats were decapitated and brain tissue was collected. Thehistological morphlogical change was investigated. Cerebral edema formation wasdetermined by brain water content using MRI methods. The expression of AQP4inbrain tissue was observed by immunohistochemistry and Real-time PCR.Results1. amlodipine treatment group MRI detected values are displayed,cerebral hematoma, edema, the CCB group and the ICCB group was better than nointervention group, and the ICCB group is superior than the CCB group, thedifference was statistically significant.(F=16.987,35.448,37.174P <0.05); Systolicblood pressure reduction ranges and reduction of the rate of edema around thehematoma has a correlation (r=0.83,0.89,0.83,p=0.000)). Diastolic blood pressurereduction ranges and reduction the rate of edema around the hematoma has a correlation (r=0.82,0.89,0.84,p=0.000).2.Down the hematoma next to edema inAQP4expression in the first days3,5,7,the CCBgroup and the ICCB group wasbetter than the untreated group, and the ICCB group is superior to the CCB group, thedifference was statistically significant.(F=217.058,21.000,51.706,P <0.05).Conclusion1.Amlodipine effective blood pressure can narrow intracerebralhemorrhage surrounding edema.2.Amlodipine by inhibiting AQP4expressionprompted hematoma surrounding brain edema.3. The increase of Systolic bloodpressure or diastolic blood pressure can promote the hematoma and surroundingcerebral edema expand.
