OBJECTIVE:1. To research the expression and clinical pathological features relationship of Livin and Caspase-3in laryngeal cancer、carcinoma and normal tissues, and to analyze the dependency between the both from the gene and protein levels.2. To explore the interaction mechanism of Livin and Caspase-3during the apoptosis of the tumor cell. By selected cases for2010-2011Shanxi Medical University first hospital and the Shanxi province tumor hospital records information full confirmed for laryngeal cancer patients, glottis Shang type30cases, glottis type16cases; clinical staging:T1period7cases, t period10cases, T3period15cases, T4period14cases; by pathology differentiation degree:high differentiation13cases, in the differentiation18cases, low differentiation15cases; there are lymph nodes transfer of25cases, no lymph nodes transfer of21cases.METHODS:1. Selecting46cases of laryngeal carcinoma and adjacent tissues of8cases and normal tissues of30cases, we are adopt SP immunohistochemical to exam the expression of Livin and Caspase-3, and combine with clinical, and analyze its relation with clinical pathological parameters.2. Selecting46cases of laryngeal carcinoma and adjacent tissues of8cases and normal tissues of30cases, we use the reverse transcriptase PCR (RT-PCR) method for detection of expression of Livin and Caspase-3.RESULTS:1. Immunohistochemical detection results:Livin and Caspase-3protein are expressed in the cytoplasm of the cell. In laryngeal cancer, and tumor-adjacent tissue and the normal tissue the positive expression rates of Livin turn lower, respectively for63.04%, and36.67%, and12.5%, by Chi square test on between the groups comparison is displayed, laryngeal cancer and other two group has significantly statistics differences (P<0.01), but the latter comparison has no clear statistics differences (P>0.05); In laryngeal carcinoma and paracancerous tissue and normal tissue the positive expression rates of Caspase-3turn higher, namely,28.26%,40.00%,75.00%, the same comparison between the groups using Chi square test is displayed, there is a significant statistical difference (P<0.01), the former two has no significant statistical differences (P>0.05). The protein expressions of Livin and Caspase-3in laryngeal carcinoma have correlation with the clinical stage, pathological grading of tumors and lymph node metastasis (P<0.05), and not associated with age, gender and site of tumor (P>0.05).2. RT-PCR results:The expression strength of LivinmRNA in laryngeal cancer, and the tumor-adjacent tissue and the normal tissue turns weakened, and is measured by the average light density value (IOD), respectively0.2807±0.0524, and0.0832±0.0332, and0.0754±0.0235, by variance analysis, the comparison between the laryngeal cancer tissue and other two groups has clear statistical differences (P<0.01), but the comparison of the latter two groups has no differences (P>0.05), the expression of Caspase-3mRNA in laryngeal carcinoma and paracancerous tissues and normal tissue follows by enhanced and is measured by IOD, respectively0.1053±0.0277,0.0277±0.0552,±0.0431, by analysis of variance, the comparison result between the laryngeal carcinoma and the other two groups has significantly statistical differences (P<0.01), the latter groups has no difference (P>0.05). The gene expressions of Livin and Caspase-3in laryngeal carcinoma have correlation with the clinical stage, pathological grading of tumors and lymph node metastasis (P<0.05), and are not associated with age, gender and site of tumor (P>0.05).3. The relational coefficient of Livin and Caspase-3by immunohistochemical method is r=-0.767, the relation coefficient measured by RT-PCR is r=-0.876. The result shows that there is a negative correlation between Livin and Caspase-3.CONCLUSION:1. Whether protein or gene level, the expression of Livin in laryngeal carcinoma is high, and the expression of Caspase-3in laryngeal carcinoma is low.2. The expressions of Livin and Caspase-3have a significant relationship with clinical stages, pathological differentiation, with or without lymph node metastasis of the tumor.3. There is a negative correlation between Livin and Caspase-3.4. Caspase-3and Livin play an important role in the occurrence and development process of the laryngeal carcinoma.5. Livin may become potential targets of therapy.6. This experiment can also be provided as a evident for this theory of the safety margin edge5mm from cancer.
