Background and Purpose:The expression of MMP-9is regulated primarily at the transcriptional level. It has beenfound that there were many loci in the MMP-9gene. Numerous studies were about theassociation between the promoter region (C-1562T) and acute myocardial infarction (AMI).However, studies about the relation of the exon polymorphisms of MMP-9gene to AMIwere rare. This study was to examine the distribution of the R279Q and R668Qpolymorphisms of MMP-9gene in Jilin province and study Whether there would be arelationship between the two loci and AMI susceptibility.Methods:137patients diagnosed with AMI and125healthy controls from the Han population inJilin province were selected. There were74male and63female patients in AMI group, theaverage age was64.87±12.60years; there were55male and70female patients in healthycontrol group, the average age was62.63±11.19years. The diagnostic standard of AMIincluded symptoms of typical chest pain; pathologic Q waves, ST segment elevation ordepression in ECG and myocardial injury marker-troponin T values more than the99thpercentile of the maximum reference value. Polymerase chain reaction-restriction fragmentlength polymorphism (PCR-RFLP) analysis was applied to detect the R279Q and R668QSNP of MMP-9gene. The continuous variables were represented by mean±standarddeviation. Two-sample t-test (measurement data) and chi-square test (count data) were usedto compare the difference of general clinical data between the AMI group and the healthycontrol group.We calculated the alleles frequency according to the genotypes, and chi-square test wasapplied to test whether the SNPs genotype was in accordance with Hardy-weinberg balance.Logistic regression analysis-AMI as the dependent variable; age, sex, BMI, TG, TC, DM,hypertension, smoking as the independent variable-was performed to assess the relevantdegree between these risk factors and AMI.Results: 1. Distribution of the R279Q polymorphism of MMP-9gene in the AMI group and thehealthy control group: the frequencies of AA genotype were35.77%and32.80%, AGgenotype were54.74%and58.40%, GG genotype were9.49%and8.80%,AG plus GGgenotype were64.23%and67.20%. The frequencies of A allele and G allele were63.14%and62.00%,36.86%and38.00%, respectively. However, the frequencies of AG plus GGgenotype and the G allele were approximate between the AMI group and the healthy controlgroup, there was no significant difference (P=0.61and P=0.79).2. Distribution of the R668Q polymorphism of MMP-9gene in the AMI group and thehealthy control group: the frequencies of GG genotype were78.83%and82.40%, GAgenotype were20.44%and16.80%, AA genotype were0.73%and0.80%,GA plus AAgenotype were21.17%and17.6%. The frequencies of G allele and A allele were89.05%and90.80%,10.95%and9.20%, respectively. However, the frequencies of GA plus AAgenotype and the A allele were approximate between the AMI group and the healthy controlgroup, there was no significant difference (P=0.47and P=0.51).3. Multivariate logistic regression analysis indicated that the TC, DM, hypertension,smoking were risk factors for AMI (OR=1.336,2.548,4.861and10.546, respectively).Conclusion:The findings suggested that no relationship between the R279Q and R668Qpolymorphisms of MMP-9gene and AMI was found in this study. TC, DM, hypertension,smoking were independently associated with AMI in the Hans of Jilin province and smokingwas the most strongly associated with AMI.
