Objective:1.To investigate the relationship between anti-mullerian hormone (AMH) and steroid hormones in follicular fluid and explore its role in the abnormal folliculogenesis of women with PCOS.2. To investigate the association between AMH and IVF-ET.Methods:Samples from23patients with PCOS and29controls (tubal infertility) undergoing IVF-ET in Women’s Hospital School of Medicine Zhejiang University from May to September in2010were collected. The levels of AMH in follicular fluid were determined by ELISA. The levels of steroid hormones in follicular fluid (FSH、 T、 P and E2) were determined by chemiluminescence assay. The fertilization rate and assisted reproduction outcome were followed up.Results:1.In patients of PCOS, AMH level was significantly increased (4.43±0.79ng/ml vs.3.86±0.73ng/ml, P<0.05, respectively) in follicular fluid when compared with controls.2. In patients of PCOS, FSH level was significantly decreased (2.24±0.89IU/L vs.4.11±2.74IU/L, P<0.05, respectively) in follicular fluid when compared with controls.3. AMH level was not correlate to E2, P, T in both groups.4. Follicular fluid level of AMH was positively and significantly correlated to the number of retrieved oocytes (r=0.496, P=0.006) and negatively and significantly correlated to follicular fluid level of FSH (r=-0.325P=0.043) in controls but not in PCOS.5. Follicular fluid level of AMH of patients began clinical pregnancy or not in both PCOS and controls were not significantly different.Conclusions:1. Oversecreted AMH in PCOS may restrict folliculogenesis by a reduction on FSH expression and an inhibitory effect on FSH sensitivity2. AMH is not a predictor of assisted reproduction outcome in both PCOS and controls. AMH may be a good predictor of the number of oocyte and fertilization rate in normoovulatory women but not in PCOS...
