ObjectiveTo observe the gastroprotection of Sulfentanyl and(or) Pantoprazole pretreatment in rats with acute gastric mucosal lesion induced by water immersion restraint stress (WIRS) in water temperature20+1and investigate it is possible mechanismMethods30male Wistar rats, wight200-250g, were randomly divided into five groups (6rats per group), including normal rats (Group NC, n=6), WRIS control group(Group WC), sufentanyl pretreatment group(Group SP), Pantoprazole pretreatment group(Group PP).sufentanyl and Pantoprazole pretreatment group (Group SPP); After6hours, the entire gaster was cut and then we detected pH of gastric fluid, degree of injury gastricmucosa and Gastric mucosal injury index(GI). Pathologic histology were observed by optical microscope of each group andwe also detected changes in ctivity of H+-K+"ATPasea, SOD and MDA.ResultsIt did not find gaster mucosa lesion or prightness fluidify in gastral cavity that NC groups had been diet for24h to compare normal rats.(1) Behavior change:There was a excitement performance in NC group, It was to last about harf an hour of WC group in cold water restraint stress, uninterrupt vocalization, imbs, hard scratching, after6hours, they were whiteness and immobility. During and after cold water restraint stress, the rats of PP group showed a similar performance with WC group Compared with the PP group, During and after cold water restraint stress, the rats of SP group and SPP group elatively quiet, occasionally limbs up and down struggle, stress6h after the performance similar to the PP group;(2) Stomach gross morphological observation:Compared with NC group, it could be seen a large number of brown liquid in the gastric lumen of rats of the WC group after WIRS6hours, and in the mucosal surface with a large number of dark red blood scab, wipe the crust of blood in gastric mucosa visible point splinter hemorrhages, and even erosion necrosis. Compared with the WC group, gastric mucosal injury induced in PP group,SPgroup and SPP group was significantly reduced, Different from PP group, he SP group had a small amount of bleeding and erosion of gastric mucosa.(3) Organization wet and dry ratio:After WIRS6hours, compared with NC group, the WC rats organization of wet and dry ratio was significantly lower (P<0.01); The difference between SPP and NC group not statistically significant (P>0.05). With WC group, the PP group, the SP group, the SPP rats organization ofwet and dry ratio was significantly increased, the PP group and the SP group, the increased statistical significance (P<0.05)(4) The difference of PH and Gastric mucosal lesion index and H+-K+-ATPase:the H+K+ATPase changes:compared with NC group, After WIRS6hours, the pH values of WC group were significantly lower, the GI values and H+-K+-ATPase activity were significantly higher (P<0.01), the PP group and the SPP group could significantly reduce water immersion restraint stress in rats the GI and H+-K+"ATPase activity (P<0.01), to raise the pH; PP group and SP group could increase the pH and reduce stress in rats UI, the H+K+-ATPase activity (P<0.01), but PP group and the result was not significant.(5) The activity of SOD and MDA to change:There were significantly lower valuer of SODand higher valuer of MDA in Group NC and WC; There were significantly higher valuer of SOD and lower valuer of MDA in Group SP and SPP; there have no statistically significant between group PP and WC.(6) Correlation analysis showed there was a negative correlation between the GI and PH and Organization wet and dry ratio and SOD, there was a positive correlation between H+-K+-ATPase and MDA; there was a negative correlation between PH and Organization wet and dry ratio; H+-K+-ATPase and SOD and MDA have a less correlation, and the expression of tissues SOD and MDA have a negative correlation.Cone l us i onThis experiment by copying immersion restraint stress (WIRS) model of rats was measured medication before and after the general extent of gastric mucosal injury and histological changes observed under light microscope, the following conclusions are the gastric pH, activity changes of H+’K+-ATPase, SOD and MDA.(1) Further evidence of sufentanil through oxygen free radicals, increased SOD activity and lower MDA activity to play a gastric mucosal protective effect; Pantoprazole through inhibition of H+-K+-ATPase activity play a gastric mucosal protective effect.(2) Card Mingshu fentanyl can reduce H+-K+-ATPase but reduce it is not obvious, pantoprazole enhance the SOD activity and reduced MDA activity. Sufentanil combined with pantoprazole does not increase SOD activity and lower MDA activity than the single effect more pronounced, indicating that pantoprazole does not play a gastric mucosal protective effect of oxygen free radicals role(3) Sufentanil combined with pantoprazole is better than the separate use of more effective inhibition of gastric acid, reducing acid-mediated gastric mucosal injury, and the combination of repulsion...
