Objective To observe the expression of the P-JAK2and P-STAT3protein in the Severe traumatic brain injury (STBI) rats established by the method of the Feeney free fall mode method.And to study the effect of radix achyranthis bidentatae decoction in the STBI rats.Methods The models of STBI rats were performed by the Feeney free falling model.Adult male Sprague-dawley(SD) rats were randomly divided into fourgroups:sham group,STBI group,radix achyranthis bidentatae group(STBI+radix),saline group,Each group had15animals. Immunohistochemistry analysis was used to examine the expression of the P-JAK2and P-STAT3protein.Moreover,the neurological recovery of rats were assessed by neurological severity scores(NSS). Results The results showed that the NSS of the STBI rats were significant differences,and the highest was the rats that were injured at24h. Furthermore, could radix achyranthis bidentatae greatly improve the STBI rats neural functional defect symptoms at168h(P<0.05).And immunohistochemistry analysis also reseached that little P-JAK2and P-STAT3protein was detected in the sham group.And compared with the sham group,there was an amount of P-JAK2and P-STAT3protein expressed in the STBI group,peaked at24h and thereafter decreased slightly(P<0.05). The expression of the P-JAK2and P-STAT3protein was lower in the STBI+radix than the model and saline groups’, especially at168h(P<0.05).Conclusion The P-JAK2and P-STAT3protein was expressed excessively and aggravating neurologic deficits symptoms of the STBI rats.And the radix achyranthis bidentatae could inhibite the expression of the P-JAK2and P-STAT3protein in the STBI rats,blocke the intracellular signalling pathway and reduce the inflammation of brain oxidative stress and cell apoptosis in the STBI rats so as to alleviate nerve function defect symptoms of the STBI rats.
