Objective:To investigate the effect of photodynamic therapy (PDT) with5-amindevullinic acid(5-AIA) as a photosenditizer on laryngeal papilloma.Methods:The effect of5-AIA-PDT on subcutaneously implanted laryngeal papillma tumor in BALB/c nude mice was studied. To establish a transplanted tumor model of human laryngeal papilloma in nude mouse.5-ALA at a dose of100mg/Kg was given intratumorally, and3h later tumors were subjected to laser irradiation (the topical application of wavelength630nm10minytes up to a total dose of90J/C m2). We set4groups (no photosensitizers or no irradiation or neither or both) to be the control groups. During the experiment,tumor sizes were measured everyday and relative tumor proliferation rates were calculated. The apoptosis index(AI) was assessed by TUNEL. Apoptosis of its Tumor Cells were observed and detected with transmission electron microscopy.The DNA contents of HPV6/11using Fluorescence quantitative PCR were measured.Results:We successfully established a human laryngeal papilloma transplanted tumor model with operation tumor samples. Tumor volume of ALA-PDT treated group was markedly smaller than the control groups.The results of transmission electroscope and in situ end-labeling of DNA breaks confirmed that5-ALA-PDT induced apoptosis.Relative tumor proliferation rates24h and48h were11.6%,17.2%,P<0.05.Conclusion:5-AIA-PDT had obvious inhibition effect on laryngeal papilloma and human papilloma virus.
