OBJECTIVE:Extract Ginger essential oil from ginger of Guangxi production,to carry on quantity-determination of the Ginger essential oil and syudy it on promoting blood circulation and removing blood stasis.Through research, clarify role of ginger essential oil on promoting blood circulation and removing blood stasis,to provide basis on retionale and experiment for the future development of Ginger essential oils.METHODS:Taking the ginger collected from Guangxi Mashan County as raw material,the essential oil was extracted by SFE.The methods of quatity-determination on Ginger essential oil with GC-MS/SIM was established. The model of acute blood stasis was established with high-molecular weight dextran in rats with drug prevention ways and the hemorheological parameters、PT and APTT of the model were observed, by which the improvement effect on model of acute blood stasis can be explained. The model of auricle microcirculatory disturbance was established with high-molecular weight dextran in mice and to observe the effect of Ginger essential oil on model.To study the effect of Ginger essential oil on promoting blood ciculation and removing blood stasis in rats comprehensively by the above research.RESULTS:It shows that the extraction rate of Ginger essential oil reached 12.8%by SFE. The GC-MS/SIM method is sensitive、high selective、accurate; in the stability experiment, the sample solution of α-Pinene、Eucalyptol and Borneol were stable in24hours. Promoting blood ciculation and removing blood stasis in pharmacological experiment shows that:①Ginger essential oil could significantly ameliorate the hemorheology and inhibit platelet aggregation in acute blood-stasis rats, it could markedly decrease the whole blood viscosity, plasma viscosity, blood reduce viscosity, hematocrit and erythrocyte conglomeration index; It could also rise erythrocyte deformation index.②Ginger essential oil could inhibit endogenous and exogenous coagulation pathway in blood-stasis rats,and could ameliorate high coagulation state in blood-stasis rats.③Ginger essential oil could against the microcirculatory disturbance induced by high-molecular dextran.CONCLUSION:The extraction method of Ginger essential oil has high efficiency,no chemicalpollution,simple process,low energy consumption; As the components of Ginger essential oil is complex,the only three components α-Pinene、Eucalyptol and Borneol are carried on quantity-determination,in order to control the quality of Ginger essential oil better,it also need futher analysis; Ginger essential oil has a good effect in promoting blood ciculation and removing blood stasis.
