Obiective Establish the model of tran-splanted endometrial cancer, adapt to different-dose Alarelin to treat,to study if Alarelin can inhibit nude mice bearing human endometrial carcinoma and to explore its possible molecular mechanisms.Methods Human endometrial carcinoma hec-l-b cells were injected into the lefe axillary fossa of the nude mousse to establish the model of tran-splanted endometrial cancer. Then the mice were randomly divided into four groups,which include low-dose group (15ug/kg), middle-dose group (30μg/kg),high-dose group (60μg/kg) and blank control group (0μg/kg). The different-dose Alarelin were injected into the kinds of mice of groups and the blank control group were injected with the isodose isotonic Na chloride in the same way. After observing the systemic condition of the mice for4weeks, the changes of xenograft morphology were observed by light microscopy with HE staining.We measured the sizes, drawed the growth curve, calculated the inhibition rate of the tumor,then, etected the expression the FHIT with the immunohistochemical technology.Results①The inhibition rate of the tumor increased obviously with the rising of Alarelin concentrations. Alarelin15、30、60μg/kg inhibted tumor by17.0%、24.1%42.1%,respectively.Compared with the control group,the difference was significant (P <0.05respectively).②The xenograft of Alarelin group showed significant necrosis morphology changes under a light microscope.③The immunohistochemical results showed that the expression of FHIT rised obviously in treated groups with the increasing dose of Alarelin. Compared with the saline group there were the statistically significant differences (P<0.05).Conclusion Alarelin might inhibit directly the growth of the human endometrial carcinoma in nude mice of subcutaneous xenograft and the effect was dose-dependent. And it may be related to the up-regulation of FHIT protein.
