Objective To exploer the effect of intravitreal injection of octreotide inhibit oxygen-induced retinopathy (OIR) mice model and investigate its mechanism.Methods Twenty-four healthy Kunming mice of7days were randomized to two groups,Normal group (group A) were raised in normal air invironment, they were raised for days, hperoxygen group were raised in airtight oxygen box for5days, they were randomized to two groups, experimental group (group B) and the control group (group C),when mice age of12days, group B octreotide3μl intravitreal injection, A group and the group C amount physiological saline intravitreal injection, afer given medicine7days, mice of19days were put to death,made eyeballs specimen. spread the retina and observed retinal vascular form under light-microscopy,HE dyed count in the film industry breakthrough vascular endothelial nuclei averages,immunohistochemical quantitative analysis expression of VEGF and IGF-1in the retina. Statistical results to mean differences and the standard deviation, SPSS13.00statistical analysis,variance analysis,compares the two groups of sample LSD-t test, P<0.05think differences with a statistical significance.Results Spread of the retina, group A of retinal vascular evenly distributed, the caliber of thicker, branch was good, peripheral retinal vascular network structure was clear, group B retinal vessels uneven distribution,part of vessels was rigid, part of pipe diameter was thinner, part of vascular around the peripapillary was blocked, there was non-perfusion area, small branch vascular of peripheral retinal was narrow, no blocked, group C distribution of retinal vessels were uneven,vessels were rigid,pipe diameter was thinner, vascular around the peripapillary was blocked, see large area without perfusion, small branch vascular of peripheral retinal was narrow and blocked, there was more non-perfusion area.group A、group B、group C HE dyeing reveal breakthrough retina world of film of vascular endothelial nuclei average trend had increased, separately as (0.27±0.23),(1.77±0.38) and (3.57±0.71) per HP,Between each group P<0.05,group A,group B,group C positive expression of VEGF in retina had increase trends,separately as (160.82±5.90),(127.14±4.40),(104.22±3.18),Between each group P<0.05, group A,group B,group C positive expression of IGF-1in retina had increase trends,separately as(145.23±5.58)、(126.65±5.13)、(105.14±4.49) expression of VEGF and IGF-1in retinal had a significantly positive correlation (P<0.05).Couclusion octreotide can effectively restrain the neovascularizationin in retinal, at the same time, reduce expression level of VEGF and IGF-1in the retina.
