In recent years, with the progress and development of modern analysis technology, Ganoderma fingerprint has become the most popular area of the traditional Chinese medicine research as the identification of the current Chinese medicine, an effective means of quality control of TCM (Traditional Chinese Medicine). IR (Infrared spectroscopy) is an important traditional Chinese medicine fingerprint; it is the identification of the compound molecules, it more specific than the qualitative identification of simple chemical functional groups, And the IR can reflect a variety of traditional Chinese medicine in the chemical composition differences. The pharmacopeia of China, UK and many other country are used the standard spectrum which prepared under extremely strict conditions as a standard, it is to say identify the drug with the standard pattern.This presents the concept of the SIR (similar infrared spectrum) combination of infrared spectroscopy and pattern recognition technology, and design the analysis software of the similar infrared spectrum. The SIR can provide the range of wave number, or location information of the most prominent difference in two IR, this can assist the spectra analysis. And it can realize the little differences between two spectra, combined with pattern recognition technology. The author analysis93kinds of Ganoderma samples in K-means Clustering method, according to the information of them IR, and the Classification results are consistent with the results of morphological classification; Establish the IR database of59kinds of Ganoderma lucidum, In the case of no a priori knowledge, can realize the automatically retrieve and intelligent recognition of Ganoderma lucidum species, according to the Ganoderma fingerprint; And proposed a new method of determination of Ganoderma lucidum polysaccharides content using fluorescence spectrometry, Can be used for the analysis of the contents of polysaccharide of the Ganoderma lucidum and other products. The specific work can be divided into the following four parts: 1. Based on comparison thought of infrared spectrum, combined with similarity calculation method in pattern recognition technology, design the analysis software of the similar infrared spectrum using the Visual Basic.net language. This software can browse infrared spectrum、calculate similar infrared spectrum of any two similar spectra、calculate different comparison precision of similar infrared spectrum by setting different offset, can calculate related coefficient and similar area of any two similar spectra between the rang of spectrum which by your setting, and it can output similar spectrum and correlation calculation data in TXT format, for its further analysis.2. Collection31samples of5kinds of Ganoderma (Ganoderma lucidum、 Ganoderma sinensis、Ganoderma atrum、Ganoderma multiplicatum and Ganoderma green), Set the first sample as the public compare object and offset is20, and calculate the similar infrared spectrum in800-1800cm-1of each samples. In1560-1502cm-1,1460-1421cm-1and1319-1260cm-1, Five Ganoderma lucidum have their own different variation, with this law, calculate all samples’correlation coefficient (R1, R2and R3) in the3Wave number range, In the case of no a priori knowledge, The K-means clustering was successfully divided them into4classes (Ganoderma lucidum and Ganoderma sinensis, Ganoderma atrum, Ganoderma multiplicatum, Ganoderma green), This result is generally consistent with the result of morphological classification。3. According to the demand analysis and retrieval method, establish the Ganoderma similar infrared spectrum database, the database has keyword queries and similarity search function. Keyword query used is the fuzzy query technology, it is more effective, less memory occupy, more effective user interaction, more strong adaptability than the traditional field queries. It can assist the people to analysis the infrared spectrum, and it can achieve material classification and quality control more convenient.4. Reference to the phenol-sulfuric acid method, Make Ganoderma lucidum polysaccharides dehydration generated furfural or hydroxyl methyl furfural under the action of the strong sulfuric acid, and then reaction whit phenol and produce a kind of benzene rings with fluorescent substance, thus we can analysis its content by fluorescence spectroscopy. We analysis9kinds of Ganoderma polysaccharide content, The results show that:It was consistent to the results of Pharmacopoeia of the People’s Republic of China. The reproducibility was good, and it provides a new measuring method of Ganoderma polysaccharide content.
