Ganoderma lucidum(Leyss.Ex Fr.Krast)is a valuable medicinal fungus with a wide range of pharmacological activity.In recent years,it has done a lot of research on its deep fermentation products such as triterpenes,polysaccharides and laccases.Due to the importance of bacteria in practice and production,it is particularly important to develop and tap new heavy resources.The pathogen of Ganoderma lucidum was studied and identified by 18S strain.The activity and application of extracellular laccase from mycelia fermentation were studied.The content of triterpenoids and polysaccharides in the fermentation broth was increased and the yield of triterpenes and polysaccharides was increased by a certain method.The main contents and results are as follows:1,The germplasm of Ganoderma lucidum was isolated and identified by tissue separation method.The mycelial DNA was extracted and identified by 18S strain.The phylogenetic tree was constructed by using the 18S sequence of the strain and the 18S sequence in the gene pool.Ganoderma lucidum and the gene pool of Ganoderma lucidum AY705969.1 sequence similar to 99%,the initial identification of the fungus Ganoderma lucidum strains,named Lushan Ganoderma lucidum.2,The results showed that the content of neutral triterpenes was higher than that of acidic triterpenes.The results showed that the content of triterpenoids was higher than that of acid triterpenes.It was found that the method of standing and standing after the first vibration was more favorable for triterpenoid synthesis by standing and shaking culture.The contents of total triterpenoids,neutral triterpenes and acid triterpenes in Ganoderma lucidum were 57.5 mg/g,53.2 mg/g and 11.06 mg/g,respectively.The total triterpenes,neutral triterpenes and acid tris The yield of total triterpenes and neutral triterpenes increased by 14%and 12.7%,respectively,and the total triterpenes and neutral triterpenes were increased by 93%,86.2%and 45.8%respectively.The strain was confirmed to be a strain of high yield neutral triterpenoids.3,Study on the active components of Ganoderma lucidum fermentation broth:we studied the contents of laccase,polysaccharide and triterpenoids in fermentation broth.Under the condition of basal culture,the laccase production of the strain was 108U/mL,and the laccase production was increased by about 3 times,up to 328U/ml by adding bran in the culture medium.In specialized media,laccase production was U/ml.Then,according to the degradation of dyes by laccase,the decolorization effect of laccase on three different dyes was explored.It was found that laccase had some decolorization effect on these dyes.Under certain conditions,crystal violet,carbolic acid and peacock The decolorization effect of stone green reached 84.4%,50.2%and 62.1%.And the highest value of polysaccharides and triterpenes in fermentation broth was 0.6659 mg/mL and 0.3243 mg/mL,further addition of the fermentation broth to nude mice,initially confirmed the strain on the tumor significantly inhibited.
