Ganoderma lucidum has a long history as one of the traditional rare medicinal herbs. It contains a great deal of constituents and pharmacological activities either it can be used in medicine or food for people. The research has made a text on activity of biological to extract from the spore powder and Ganoderma lucidum. The experiment results are as follows:(1) Study on water-soluble polysaccharides determined by central composite design.We make an investigation to extract between the spore powder and Ganoderma lucidum in the same temperature, extraction time and solid-to-liquid ratio. We have found the best extraction condition according to ridge analysis was as follow:the temperature was 99 degree, the extraction time was 197 min, the solid-to-liquid ratio was 135mL/g by central composite design and response surface methodology. And the best extraction condition of Ganoderma lucidum was as follow:the temperature was 98 degree, the extraction time was 206 min, the solid-to-liquid ratio was 80mL/g.(2) Study on water-soluble protein determined by central composite design.We make the same investigation to extract between the spore powder and Ganoderma lucidum in the same temperature, extraction time and solid-to-liquid ratio. We have found the best extraction condition according to ridge analysis was as follow: the temperature was 94 degree, the extraction time was 157 min, the solid-to-liquid ratio was 166mL/g by central composite design and response surface methodology. And the best extraction condition of Ganoderma lucidum was as follow: the temperature was 92 degree, the extraction time was 195 min, the solid-to-liquid ratio was 173mL/g.(3) Study on Ganoderic acid A determined by central composite design.We make the same investigation to extract between the spore powder and Ganoderma lucidum in the same ethanol concentration, extraction time and solid-to-liquid ratio. We have build ultrasonic extraction and the mathematical model of Ganoderic acid A rate by central composite design and response surface methodology. So that we have get the precision of regression equation coefficient is 0.9615 and the equation is fitted. And the best extraction condition according to ridge analysis was as follow: the ethanol concentration was 70%, the extraction time was 41 min, the solid-to-liquid ratio was 82mL/g by central composite design and response surface methodology. The best extraction condition of spore powder was 0.9565 and the equation was fitted, too. The ethanol concentration was 49%, the extraction time was 40 min, and the solid-to-liquid ratio was 60mL/g.(4)Study on Ganoderic acid A by enzymatic hydrolysisThe best extraction technology for Ganoderma lucidum was 1% laccase, 3% pectinase, 5% mannanase, 30 min, 40℃,pH6. Under this condition the extraction rate of Ganoderic acid A was 3.521%. On the other hand, the best technology for the spore powder was 1% laccase,3% glycosidase, 30 min, 40℃, pH6, and the rate of Ganoderic acid A was 0.5422%.(5) Study on pharmacokinetics of Ganoderic acid A.The object of this study was the pharmacokinetics parameters of Ganoderic acid A dissolved in 60% alcohol and water.There was three dose groups: 184mg/kg, 920mg/kg, 1840mg/kg.Plasma concentration-time curves were charted by the 3P97 pharmacokinetics fitting procedures.The results showed that the pharmacokinetic model was 2 chamber model.From the half-life T1/2beta and the period area under the curve of standard,we found there was a linear relationship between AUC and the dose.We found that the pharmacokinetics parameters of alcohol solution was better than water solution,so it has better bioavailability.
