Objective:To assess the effects of SZ-125on coronary arterial thrombosis in the Macaca mulatta model of Femoral artery Balloon Injury.Methods:30macaca mulattas were randomly divided into5groups according to the administration of different antiplatelet drugs. The first group(n=18) macaca mulattas took SZ-125,6given0.1mg/kg,6given0.2mg/kg and6given0.3mg/kg. The second group(n=6) macaca mulattas received a comparable volume of Saline. The third group(n=6) macaca mulattas received a bolus of14μg/kg Tirofiban. The plasma levels of Antigen of vWF、the functional binding of vWF to collage、the SZ-125and vWF occupancy by SZ-125were determined using a sandwich ELISA as described.30macaca mulattas were followed up for Intravascular thrombosis for48h.Results:(1)The bolus injection of SZ-125resulted in a full occupancy of vWF by SZ-125that was sustained for at least4h.(2)the ex-vivo binding of vWF to collagen was completely blocked for4h after the antibody administration.(3)vWF:Ag levels were not affected by the SZ-125treatment throughout the experiment.vWF:Ag levels and vWF:CBA were normal in control macaca mulattas and macaca mulattas received Tirofiban.(4) Administration of SZ-125did not result in thrombocytopenia and was not accompanied with a significant prolongation of the bleeding time.Conclusion:Administration of SZ-125resulted in a complete inhibition of vWF binding to collagen and platelet adhesion. It will be a safe and feasible medicament in the prevention of in-stent restenosis.
