Objective:To establish the Lewis rat subcutaneous endometriosis model, and observe pathology, investigate how expression of mRNAs of endometrium estrogen receptor (ER) and progesterone receptor (PR) contribute to endometriosis.Methods:Taking Lewis rat as a donor, endometriums were subcutaneous injected into28rats.7days and21days after surgery heterotopic nodules were removed and stained by HE. The pathology of heterotopic nodules was observed. The expression of mRNAs of endometrial estrogen receptor (ER) and progesterone receptor (PR) were detected by reverse transcription polymerase chain reaction (RT-PCR); Differential expression of ER, PR and ER/PR mRNA changes of7days group and21days group and orthopotic endometrium group were analyzed.Results:1. All rats of this experiment survived. Survival percentage of ectopic endometrium planted in abdominal wall was100%, which was the raised small cystic mass in the abdominal wall and with the basic organizational structure of the normal endometrium.2. Volume of heterotopic nodules of21days group were smaller than7days group, the difference being statistically significant (P<0.01).3. ER and PR mRNA expression of7days group and21days group is lower than ectopic endometrium group, the difference being statistically significant (P<0.01). There is no significant difference between7days Group and21days group in ER, PR mRNA expression.4. ER/PR mRNA of7days group and21days group is higher than orthopotic endometrium group, the difference being statistically significant (P<0.01). There is no significant difference between7days Group and21days group in ER/PR mRNA ratio.Conclusions:1. Subcutaneous injection of endometrium in Lewis rats abdominal wall to establish endometriosis model has many advantages, such as simplicity, intuition, slight wound, high success rate, the same physiological characteristics of ectopic endometrium and entopic endometrium, facility to repeated Study and continuous observation.2. There is relevant between the decrease of ER of ectopic endometrium and occurrence and development of endometriosis estrous cycle inhibited of rats.3. There is relevant between the increase of ER/PR of ectopic endometrium and occurrence and development of endometriosis estrous cycle inhibited of rats.
