Abstract/Summary:
Objective:1the study of the main Pharmacodynamics of Changji’an capsule(1) Changji’an capsule affected the advanced rate of the rat’s small intestine、the rat’s small intestine which was influenced by Neostigmine、 the rat’s small intestine which was influenced by Atropine;(2) Studying of function of antidiarrheal、analgesic effect of Changji’ an capsule;(3) Studying of pain, diarrhea、unadjusted liver and spleen IBS rat model which was influenced by Changji’an capsule;2Toxicologic studies of Changji’an capsule(1) Studying acute toxicity of Changji’an capsule to evaluate toxicity rating and the safe volune of clinical use;(2) Studying long-term toxicity of Changji’an capsule to get range of safe dose.Methods:1the main Pharmacodynamics of Changji’an capsule(1)Changji’an capsule affected the advanced rate of the rat’s small intestine, mainly included three aspects:advanced rate of the rat’s small intestine、of the rat’s small intestine which was influenced by Neostigmine、 of the rat’s small intestine which was influenced by Atropine;(2)Changji’an capsule affected on the mice in diarrhea which was induced by Senna. Making the model of diarrhea to study the efficacy of Changji’an capsule; (3)Changji’an capsule affected on the pain rat which was induced by hot plate(4)Changji’an capsule effected on the pain and lax IBS rats;(5)Changji’an capsule effect on unadjusted liver and spleen IBS rats.2Toxicologic studies of Changji’an capsule(1)Acute Toxicity study of Changji’an capsule:using the mice’s maximum tolerated dose to study its acute toxicity;(2)Extended toxicity study of Changji’an capsule:Per kilogram rat was given high dose of Changji’an capsule15.624g crude drug (60times of ault’s clinical dose)、middle dose of Changji’an capsule7.812g crude drug (30times of ault’s clinical dose)、 low dose of Changji’ an capsule3.906g crude drug (15times of ault’s clinical dose) everyday, continous administration for six weeks, withdrawal recovery for two weeks. All indices of after administration compared with the blank group such as general status、body weight、food intake、water intake、urine、hematology、blood chemistry、organ coefficient histopathology and so on.Results:1Changji’an capsule (8.12、4.062、2.031、1.016、0.508g crude drug·kg-1) five dose all can restrain the advanced rate of the rat’s small intestine (P<0.01)、of the rat’s small intestine which was influenced by Neostigmine (P<0.01)、of the rat’s small intestine which was influenced by Atropine (P<0.01)2The result showed that Changji’an capsule (4.062、2.03、1.016g crude drug·kg-1) three dose all can reduce stool points of0-3h、3~6h, and had obvious dose-effect relationship;3The result showed that Changji’an capsule (4.062、2.031、1.016g crude drug·kg-1) three dose all can obviously increase pain threshold of rats (P<0.01or P<0.05)4The result showed that Changji’an capsule (4.062、2.031、1.016g crude drug·kg-1) three dose all can increase the body of mice (P<0.01or P<0.05) decrease the wet weight of rats stool (P<0.01)、SS、VIP of mice;5The result showed that Changji’an capsule (4.062、2.031、1.016g crude drug·kg-1) three dose all can increase the threshold of abdomen lifted and extrodos of mice (P<0.01or P<0.05)、increase5-HT of mice; 6Result of the study of acute toxicity showed that activities、eating、 drinking、weigh、urin、blood chemistry、coefficient of organ and so on of rats were normol;7All indices of after administration for six weeks compared with the blank group such as general status、body weight、food intake、water intake、urine、 hematology、blood chemistry、organ coefficient histopathology and so on, we can’t see toxic sensitive indice and organ;All indices of high、middle、low dose of Changji’an capsule were normal after withdrawal recovery, we can’t see delayed and accumulated toxicity.Conclusion:Changji’an capsule had the treatment of the pain and lax IBS rats、 unadjusted liver and spleen IBS rats; had the inhibition of advance rate of the intestine of rats, and had the effect of antidiarrheal、pain reliving; the clinical dose of ault and the duration of treatment about Changji’an capsule were both in a safe range.
