Croton is a genus of Euphorbiaceae comprising around1,300species, which is wide spread in tropical regions. Several species have a long role in the traditional use of medicinal plants in Africa, Asia and South America. Popular uses include treatment of cancer, constipation, diabetes, digestiveproblems, dysentery, externalwounds, fever, hypercholesterolemia, hypertension inflammation, intestinal worms, malaria, pain, and ulcers. Diterpenes are very common in Croton, a wide diversity of effects having been noticed, including anti-cancer, ant-pathogenic microorganisms, antioxidant, anti-inflammation analgesic, hypolipidemic. The recent pharmacological studies showed that medicinal croton plants have multiple bioactivities, especially the activities of antibacterial, antivirus and anti-tumor, which attracted more and more attention. In fact, few researches have been performed on these medicinal plants in china, and the researches of croton chemical constituents mainly due to foreign. So it is necessary and meaningful to do some research works to develop the abundant resources of medicinal croton plants in our country.Croton crassifolius Geisel belongs to the genus croton of the family euphorbiaceae. Distributed mainly in the south of china, such as the provinces of hai nan, guangdong, guangxi and fujian. The plant has been used as a folk medicine to treat rheumatoid pain, low back pain, stomach pain, sore throat, jaundice, colic, snake bites. We examined the80%methanol extract of the roots of this plant through various chromatographic techniques, and elucidating their structures on the basis of physic-chemical property and spectroscopic, and determined the main constituent of cyperenoic acid by HPLC analysis.In our systematical study on the chemical constituents of Croton crassifolius Geisel,19compounds were obtained from the80%methanol extract of the roots of this plant through various chromatographic techniques(silica gel, Sephadex LH-20and RP C18).15of their structures were elucidated on the basis of physic-chemical property and spectroscopic analysis (IR, UV, MS,1H and13C NMR, HSQC, HMBC,1H-1H COSY and NOESY). The15compounds from Croton crassifolius Geisel are5(10),13E-ent-halimandien-15,16-olide-19α-oicacid(1),5(10),13E-ent-halimandien-15,16-olide-19α-oic acid methyl ester(2), isoteufin(3), teucvidin(4), teucvin(5), chettaphanin Ⅰ (6), mallotucin C(7), mallotucinD(8),(12S)-15,16-epoxy-6β-methyoxy-19-norneoclerodane-4.13(16),14-triene-18,6α,20,12-diolide(9), cyperenol(10), cyperenoic acid(11), acetyl aleuritolic acid(12), lupeol(13),β-sitosterol(14) and stigmasterol(15). Nine of the compounds are diterpenoids. and two sesquiterpenes, two triterpenoids, two steroids.compounds1and2are new and novel compounds, according to the best of our knowledge. Compounds3,5,7,8,9were isolated for the first time from this plant.On the basis of systematical study on the chemical constituents of Croton crassifolius Geisel, we further analyzed the constituent cyperenoic acid of Croton crassifolius Geisel by HPLC assay. the mobile phase was methanol-0.1mL/L formic acid (87:13); the detection wavelength was240nm; the flow rate was1.0mL/min. The results show that the linear range of cyperenoic acid was0.8μg~5μg. The average recovery ratio was98.54%with the relative standard deviations (RSD) of2.53%. Methodological evaluation and results show that it is a reliable approach having some advantages as convenient, efficient, sensitive and being-reproducibility. Providing a reliable method establish the quality control of Croton crassifolius Geisel.In addition, recent advances in the research of chemical constituents and pharmacological roles of croton plants were reviewed with108references.
