The plant Croton crassifolius Geisel(Euphorbiaceae) is widely distributed in south China, Thailand, Laos, Vietnam, etc. In folk, this plant is applied to treat cancer,sorethroat, stomachache and rheumatism. In our early study, the chemical constituents of the C. crassifolius were studied and several compounds exhibited the anti-angiogenic activities on Tg(fli1a:EGFP) y1-type zebrafish embryos. In order to obtain more active compounds from C. crassifolius, the chemical constituents of SFE extract from the roots of C. crassifolius were studied. Thirty-three compounds were isolated and they were identified as crassifolin J(CC-1), crassifolin K(CC-2),crassifolin L(CC-3), crassifolin M(CC-4), crassifolin N(CC-5), crassifolin O(CC-6), crotonpyrone C(CC-7), crassifolin A(CC-8), crassifolin B(CC-9),crassifolin C(CC-10), crassifolin D(CC-11), chettaphanin I(CC-12),1,4-methano-3-benzoxepin-2(1H)-one(CC-13), isoteucvin(CC-14), teucvidin(CC-15), neoclerodan-5,10-en-19,6β,20,12-diolide(CC-16), chettaphanin II(CC-17),crassifoliusin A(CC-18), penduliflaworosin(CC-19), crassifolin F(CC-20),cyperenoic acid(CC-21), cyperenol(CC-22),(4S*,7R*,8R*,10S*)-8-hydroxy-α-guaiene(CC-23),(+)-guaial(10),ll-dien-9-one(CC-24), spathulenol(CC-25), araradendrme diol(CC-26),4,5-secoguaia1(10),11-diene-4,5-dioxo(CC-27), 1β,11-dihydroxy-5-eudesmene(CC-28), crotonpyrone B(CC-29), crotonpyrone A(CC-30), acetyl aleuritolic acid(CC-31), lupenone(CC-32) and lupeol(CC-33). Among them, CC-1~CC-7 were new compounds, and CC-15, CC-23~28, CC-32 were isolated from this plant for the first time.All the compounds isolated from the SFE extract of the roots of C. crassifolius were evaluated for their anti-angiogenic activity using a wild-type zebrafish in vivo model. And, compounds CC-8, CC-9, CC-19, CC-21, CC-23 and CC-24 exhibited inhibitory activity against vessel formation on zebrafish embryos.
