Croton crassifolius Geisel is a small shrub that grows widely in the south and southwest parts of China,Vietnam,Thailand and Laos.The roots of Croton crassifolius are used as a traditional medicine for treatment of low back pain,rheumatoid pain,stomachache,sore throat,snake bites,and cancer.Caesalpinia bonduc L.Roxb.(Leguminosae),is a medicinal plant widely distributed throughout the tropics and subtropics.In traditional medicine,different parts of the plant are used to treat asthma,chronic fever,coughs,headache and stomach upset.In our systematical study on the chemical constituents of Croton crassifolius and Caesalpinia bonduc,these compounds were isolated by means of CC,TLC,PTLC and Sephadex LH-20 etc.The structures of these compounds were determined on the basis of spectroscopic analysis,such as IR,UV,MS,HR-ESI-MS,1D-NMR(’H-NMR,13C-NMR,DEPT),2D-NMR(’H-’H COSY,HSQC,HMBC,NOESY)and X-ray crystallography.Twenty-one known compounds were isolated from roots of Croton crassifolius,along with five new compounds.Eleven known compounds were isolated from seed kernels of Caesalpinia bonduc,long with two new compounds..New compounds were tested for cytotoxic activity against human cancer cell lines using the MTT method,and BYD-05 was further evaluated for antibacterial activity.Both BYD-01 and BYD-02 were evaluated for their cytotoxicity against four human cancer cell lines,HepG2,SGC-7901,K562,and K562/ADM,Unfortunately,no significant activity was detected;BYD-03 showed no cytotoxicity against tested cell lines(HepG2,SGC-7901,and K562);Compounds BYD-04 and BYD-05 exhibited good activity against two human cancer cell lines(HeLa and NCI-446)with IC50 values in the range of 6.52-10.21 μg/mL.Moreover,BYD-05 was further evaluated for antibacterial activity against two Gram-positive(Staphylococcus aureus and Enterococcus faecalis)and one Gram-negative bacteria(Escherichia coli).BYD-05 showed moderate activity against S.aureus with a MIC value of 50 μg/mL.The purpose of this paper is about the chemical composition of Croton crassifolius and Caesalpinia bonduc further research,and biological activity were studied for the part of the chemical composition,which will provide a scientific basis for traditional Chinese medica and the screen of new medicine.
