Effects Of Traditional Chinese Medicine "Bu Shen Hua Yu" Formula On Bone Biochemical Metabolism Ovariectomized Osteoporosis Rats

Objective: To prepare the formula Qiang Ku Kang Shu (QGKS) capsule forthe therapy of primary osteoprpsis based on the theory of traditional Chinesemedicine “Bu Shen Hua Yu”-tonifying the kidney and removing blood stasis tocompatibility of medicines. To evaluate the efficacy of QGKS in treatment ofosteoporosis (OP) by observation of serum BALP, TRACP, MMP-2, TGF-β1,calcium, phosphorus and bone mineral density (BMD) in ovariectomizedosteoporosis rats.Methods:(1) Through arranging documentation, generalize the theoreticalbasis of tonifying the kidney and removing blood stasis method in the treatment ofprimary osteoporosis.(2) In this experiment, forty sprague dawley(SD) rats at ageof six months were used, and were randomly divided into blank group, model group,estrogen group, high dose of traditional Chinese medicine group, as well as low doseof traditional Chinese medicine group. Except of the blank group, we used themethod of removing both ovaries in the other groups’ rats to copy osteoporosismodel. One month after the operation on rats, rats in each group were began to begavaged by the volume of2ml every day, and with the dose in each group as follows:estradiol valerate1mg/kg/d, high dose of QGKS group: QGKS capsule16g/kg/d(calculated on a crude drug, the same below), low dose of QGKS group: QGKScapsule8g/kg/d, blank group and model group were fed with saline solution.(3)After one month’s intragastric administration, blood was collected by the method ofheart blood sampling, and then separated serum from it which was used to detectionserum BALP, TRACP, MMP-2, TGF-β1, calcium and phosphorus. The right femurwas separated for BMD testing.(4) Statistical analysis: All dates were expressedwithx±s, compared the differences among the groups with T test.Results:(1) The effects on BMD: there are significant differences betweenthe blank group and the model group (P<0.01). High dose of QGKS group, estrogengroup were also different from the model group (P<0.05). The values of BMD in theestrogen group and high dose of QGKS group were lower than the blank group(P<0.05), and there is no statistical difference between the estrogen group and highdose of QGKS group (P>0.05).(2) The effects on serum BALP: The high dose ofQGKS group compared to the model group has significant differences (P<0.01). Theestrogen group and blank group were also different from the model group (P<0.05). The low dose of traditional QGKS group compared to model group has statisticalsignificance (P<0.05). Comparing with high dose of QGKS group, estrogen groupand blank group, there had no significant difference (P>0.05), but the high dose ofQGKS group seems to has a trend to be higher.(3) The effects on serum TRACP:Compared to the model group, serum TRACP in the high dose of Chinese medicinegroup were significantly reduced (P<0.05), but the blank group and estrogen group’sreduced more (P<0.01). Comparing with the high dose of QGKS group, estrogengroup and blank group, there had no significant difference (P>0.05). The low dose ofQGKS group compared to the model group had also no obvious statisticalsignificance (P>0.05).(4) The effects on serum MMP-2: Compared the estrogengroup and blank group to the model group had significant statistically(P<0.01).There are differences between the high dose of QGKS group and the modelgroup (P<0.05). Comparing with the high dose of QGKS group, estrogen group andblank group, there had no significant difference (P>0.05). The low dose of QGKSgroup compared to the model group had also no obvious statistical significance(P>0.05).(5) The effects on serum TGF-β1: Comparing with the blank group andmodel group, there was significant differences (P<0.01). The estrogen group andhigh dose of QGKS group were also different from the model group (P<0.05). Thevalues of serum TGF-β1in the high dose of QGKS group were lower than in theblank group while serum TGF-β1in the estrogen group were the same as blankgroup.(6) The effects on serum calcium and phosphorus: The changes of serumcalcium and phosphorus in each group had the same trend: The model group rats’serum calcium and phosphorus were more than the high dose of QGKS group,estrogen group and blank group (P<0.05). The low dose of QGKS group comparedto the model group was not significantly different (P>0.05). Comparing with thehigh dose of QGKS group, estrogen group, blank group had no obvious statisticalsignificance (P>0.05).Conclusions: According to the theory of “Bu Shen Hua Yu”-tonifying thekidney and removing blood stasis for therapy of primary osteoporosis,the formulaQiang Gu Kang Shu capsule was extablished. This study proves that QGKS has theeffects of promoting the BMD in ovariectomized osteoporosis rats, and improvingbone metabolism in osteoporosis rats. The evidence above confirmed that QGKScapsule may play a good therapeutic effect on primary osteoporosis.
