Objective: The aim of this paper was to investigate the expression ofStathmin, PDCD4and Caveolin-1proteins in cholangiocarcin-oma and therelation to the oncogenesis of cholangiocarcinoma.Methods: The expression of Stathmin, PDCD4and Caveolin-1proteinsin43chlolangiocarcinoma tissues and22adjacent bileduct tissues wereexamined by immunohisto-chemistry method. The relation of these threeproteins to clinical and pathological features was analyzed. Flow cytometryanalysis was employed to measure the expression of Stathmin, PDCD4andCaveolin-1proteins in22specimens of chlolangiocarcinoma tissues and22specimens of adjacent non-neoplastic tissues respectively. SPSS13.0wasapplied to analyze the results of experiment.Results:1Results of immunohistochemistryImmunohistochemistry was employed to detect the expression of Stathmin,PDCD4and Caveolin-1proteins in43specimens of chlolangiocarcinomatissues and22specimens of adjacent non-neoplastic tissues. The positiveexpression rate of Stathmin, PDCD4and Caveolin-1proteins inchlolangiocarcinoma tissues was93%(40/43),44.2%(19/43),37.2%(16/43),while that in the adjacent non-neoplastic tissues was40.9%(9/22),77.2%(17/22),13.5%(3/22), respectively. There was significant difference in thepositive rates of Stathmin, PDCD4and Caveolin-1proteins betweenchlolangiocarcinoma tissues and the adjacent non-neoplastic tissues (P<0.05).The positive expression rates of these proteins were also different in differentdifferentiation degree.2Results of Flow cytometry analysisFlow cytometry analysis was employed to detect the expression ofStathmin, PDCD4proteins in22specimens of chlolangiocarcinoma tissues and22specimens of adjacent non-neoplastic tissues. The mean level ofStathmin, PDCD4proteins was230.97±21.41and228.84±32.10inchlolangiocarcinoma tissues, while that in the adjacent non-neoplastic tissueswas152.12±20.05and293.45±25.72, respectively. There was significantdifference in the mean level of Stathmin and PDCD4proteins betweenchlolangiocarcinoma tissues and the adjacent non-neoplastic tissues (P<0.05).The mean level of Caveolin-1protein was272.76±7.57inchlolangiocarcinoma tissues, while that in the adjacent non-neoplastic tissueswas270.25±6.74.There had no significant difference in the mean level ofCaveolin-1protein between chlolangiocarcinoma tissues and the adjacentnon-neoplastic tissues (P>0.05).There was significant difference in the mean level of Stathmin andPDCD4proteins in pathological grading of tumor.(F=281.94, P=0.03<0.05;F=57.760, P=0.03<0.05, respectively).There was significant difference in the mean level of PDCD4protein inclinical stage of chlolangiocarcinoma tissues (t=-3.333, P=0.001<0.05). Themean level of PDCD4protein had no relation to lymph node metastasis(P>0.05), but the mean level of PDCD4protein in lymph node metastasisgroup (221.75±30.26) was lower than the group without lymph nodemetastasis (265.79±1.73). The mean level of Caveolin-1protein wasn’tcorrelated with clinical stage of tumor (P>0.05) and lymph node metastasis oftumor (P>0.05).3There was no relation among Stathmin, PDCD4and Caveolin-1proteinsin chlolangiocarcinoma tissues.(There was no relation between Stathmin andPDCD4proteins in chlolangiocarcinoma tissues. r=0.060, P=0.695>0.05;There was no relation between Stathmin and Caveolin-1proteins inchlolangiocarcinoma tissues. r=0.615, P=0.274>0.05There was no relationbetween PDCD4and Caveolin-1proteins in chlolangiocarcinoma tissues.r=0.273, P=0.063>0.05).Conclusions1Results of immunohistochemistry and flow cytometry analysis show that the higher expression of Stathmin protein and the lower expression of PDCD4protein are correlated with tumorigenesis and progression ofchlolangiocarcinoma tissues.2Results of immunohistochemistry and flow cytometry analysis showthat the higher expression of Stathmin protein is correlated with pathologicalgrading and the expression of Stathmin protein is correlated without lymphnode metastasis and clinical stage of chlolangiocarcinoma tissues.3Results of immunohistochemistry and flow cytometry analysis show thatthe lower expression of PDCD4protein is correlated with pathological gradingand lymph node metastasis and clinical stage of chlolangiocarcinoma tissues.And the lower expression of PDCD4protein is correlated with biologicalbehavior and prognosis of chlolangiocarcinoma.4Results of immunohistochemistry show that the higher expression ofCaveolin-1protein is correlated with pathological grading and lymph nodemetastasis and clinical stage of chlolangiocarcinoma tissues. And theexpression of Caveolin-1protein is correlated with biological behavior andprognosis. While the results of flow cytometry analysis show that theexpression of Caveolin-1protein is not correlated with pathological gradingand lymph node metastasis and clinical stage of chlolangiocarcinoma tissues.5Stathmin, PDCD4and Caveolin-1proteins maybe play an important role ina series of pathological progression (eg: tumorigenesis, progression, invasionand metastasis) of chlolangiocarcinoma tissues.
