Objective:This study is to investigate the association between femalepostmenopausal osteoporosis and serum25-OHD3and also to explore the relationshipbetween female postmenopausal osteoporosis and vitamin D receptor polymorphismon genetics and molecular biology, to provide the theory basis and evidence forprevention and treatment of osteoporosis.Methods: Serum25-OHD3of208postmenopausal women was measured by thechemiluminescence method. Observe their vitamin D and divide them into differentage groups to explore the relationship between Serum25-OHD3and ages. Bonemineral density(BMD) on lumbar spine(L1-L4) were measured by dual energyX-ray absorptiometry. The subjects were divided into osteoporosis group(n=126),andnon-osteoporosis group(n=82) according to the measuring result. Investigate theassociation between postmenopausal women’s BMD and serum25-OHD3. VDRTru9I gene polymorphism was assessed by the polymerase chain reaction(PCR)-restriction fragment length polymorphism(RFLP) method. The following step iscomparing the serum25-OHD3level, the frequency distribution of VDR Tru9I genepolymorphism and allele in two groups.Result:The serum25-OHD3level in postmenopausal osteoporotic women were(14.59±5.72)ng/ml. In non-osteoporosis group they were (18.38±7.01)ng/ml.Postmenopausal osteoporotic women had a lower concentration (P<0.05). The levelof serum25-OHD3were correlated with postmenopausal women’s T score (r=0.416,P<0.05).142(68.3%) postmenopausal women were lack of vitamin D,61(29.3%) cases were insufficient,5(2.4%) cases were sufficient. There were no correlationbetween serum25-OHD3level and ages(P>0.05). The genotype frequencydistribution of Vitamin D receptor gene followed the Hardy-Weinberg equilibrium(P>0.05). The frequency distribution of genotypes UU, Uu and uu were60.3%,35.7%and4%, the allele U78.2%, u21.8%in the osteoporosis group respectively. Inthe control group the frequency distribution of genotypes UU, Uu and uu were85.4%,13.4%and1.2%, the allele U91.5%, u8.5%. There were statistical difference of thefrequency distribution of genotypes and alleles between two groups(P<0.05). Thedistributing frequency of uu genotype and u allele in osteoporosis women was higherthan the control group.Conclusions:1.Serum25-OHD3level is associate with female postmenopausalosteoporosis, and it may be one of the importante reason for bone mass reduction inpostmenopausal women. Serum25-OHD3level is not correlate with ages, butosteoperotic degree. Most of postmenopausal women were lack of vitamin D.2. The VDR Tru9I gene polymorphism is associate with postmenopausal osteoporosisin this study. u allele is susceptive factor of postmenopausal osteoporosis. The VDRTru9I gene polymorphism site can be the genetic marker of postmenopausalosteoporosis in Yue Yang.
