[Objective]To observe effects of abdominal injection of Salvianolate and PVP-I on rats with postoperative intestinal adhesion and to explore the functioning mechanism.[Methods]88Male SD rats were randomly divided into five groups:normal control group (n=8), model control group(n=32), salvianolate group (n=8), PVP-I group (n=8) and salvianolate combined with PVP-I group(n=32).The model group and the salvianolate combined with PVP-I group respectively divide into4groups, according to1,3,5,7four time points (n=8). Except normal control group, intestinal adhesion modeling should be conducted to other rats according to Hugh. The normal control group and model control group of intraperitoneal injection of5%glucose injection4ml/kg, continuous intravenous injection for7days. Salvianolate group daily intraperitoneal injection of Salvianolate10.9mg/kg, continuous intravenous injection for7days; PVP-I group daily intraperitoneal injection of0.625%PVP-I dilution of4ml/kg, continuous intravenous injection for7days; the combination group daily intraperitoneal injection of salvianolate10.9mg/kg and PVP-I4ml/kg, continuous intravenous injection for7days. The model control group and combination group after operation respectively1,3,5,7days, other groups are respectively on the seventh postoperative day in U shaped incision open during narcosis, according to the Nair classification standard, observed and assessed intestinal adhesion grade in each group by the naked eye; Immediately, sampled, anti-coagulated and centrifugated the blood of postcava to obtain the blood serum, and preserved it at an ultra-low temperature. A later ELISA method would be employed to test the concentration of IL-1β and TGF-β1in the blood-serum samples. Sampled the adhesive tissues from the most obvious part with the size of1.0cm×1.0cm×0.5cm, and placed the samples in the10%neutral formaldehyde fixative for24hours. Embed the samples in the paraffin, made slices, conducted the HE coloration, and observed expression of fibroblast and inflammatory cells by microscope observation.[Results]After seventh days, all the rats were survived, the experimental results show.1. Intestinal adhesion:comparison with normal control group, model group animal have adhesions (P<0.05), and a higher degree of adhesion. Compared with the model group, salvianolate group, the combination group of intestinal adhesion reduced significantly (P<0.05, P<0.01); but there was no statistical significance in PVP-I group compared with the model group (P>0.05).2.Serum IL-1β, TGF-β1level comparison:model control group IL-1β, TGF β1levels were significantly higher than normal control group (P<0.01); compared with model control group, salvianolate group, PVP-I group IL-1β, TGF-β1decreased (P<0.05, P<0.01), compared with model control group combination group IL-1β, TGF-β1were significantly reduced (P<0.01).3. At each time points after operation of rat serum IL-1β、TGF-β1dynamic changes:model control group serum IL-1β on the seventh postoperative day reached the highest; the combination group IL-1β on the fifth postoperative day reached the highest, and the seventh day declined. Two groups of all time points corresponding comparison, there were significant differences in fifth,seventh2time points (P<0.05, P<0.01); the model control group TGF-β1on the seventh postoperative day achieved the highest; the combination group TGF-β1on the fifth postoperative day reached the highest, on the postoperative seventh day declined. Two groups of all time points corresponding comparison, there was significant difference in fifth,seventh2time points.(P<0.05, P<0.01).4. Light microscopic observation of rat tissues in fibroblasts and inflammatory cells and expression of basic proportional relationship. [Conclusion]PVP-I, salvianolate, PVP-I combined with salvianolate can reduce postoperative intestinal adhesion degree, and reduce the IL-1(3and TGF-β1expression levels, inhibition of fibrous connective tissue hyperplasia,and the combination group effect is more obvious; Cytokine IL-6, TNF-a in intestinal adhesion formation play a certain role...
