Objective To establish the processing technology of flowing Verbena officinalis L. yin pian; To compare the traditional Verbena officinalis L. yin pian with flowing Verbena officinalis L. yin pian; To develop the quality standard of flowing Verbena officinalis L. yin pian; To study the stability of flowing Verbena officinalis L. yin pian.Methods①Wet granulation and extrusion-spheronization were used to prepare flowing Verbena officinalis L. yin pian (pills). Sphericity, yields, Ursolic Acid&Oleanic Acid content were considered as evaluation indicators. Orthogonal design and multi-indicator score system were applied to evaluate the effects by water ratio, spheronization speed and spheronization time. The optimized processing technology parameters were obtained.②The difference between flowing Verbena officinalis L. yin pian and traditional Verbena officinalis L. yin pian was studied from the following levels:aspect, microscopic identification, TLC chromatography, content of Ursolic Acid and Oleanic Acid, HPLC chromatography, decocting rate and HPLC chromatography of decoction.③The regular quality standard items of flowing Verbena officinalis L. yin pian were tested, referring to Chinese Pharmacopoeia (2010Edition), such as aspect, microscopic identification, TLC chromatography, content of Ursolic Acid and Oleanic Acid, content of water, total ash and acid-insoluble ash, sphericity, bulk density, friability, repose angle, and simultaneous determination of Aucubin, Hastatoside, Verbenalin and Verbascoside.④The aspect, TLC chromatography and the index components content were considered as investigation indicators. Flowing Verbena officinalis L. yin pian under high temperature, high humidity, strong irradiance conditions, accelerated stability and long time stability were investigated. Traditional Verbena officinalis L. yin pian was set up as control group in the study of accelerated stability and long time stability.Results①The optimum is:80mesh powder, handling at95℃for4hours, add water ratio1:2.2, rotational speed500rpm, rotational time,20min, drying by105℃. The Flowing Yin Pian presented a perfect sphericity, uniformity size, a good yield and the content meet requirements of the pharmacopoeia.②There were no significant difference in comparison of flowing Verbena officinalis L. yin pian and traditional Verbena officinalis L. yin pian. The decocting rate of flowing Verbena officinalis L. yin pian was better than traditional Verbena officinalis L. yin pian.③The regular quality standard items of flowing Verbena officinalis L. yin pian were tested according to Chinese Pharmacopoeia (2010Edition). Besids, sphericity, bulk density, friability, repose angle and simultaneous determination of several components were also considered. The established HPLC method for simultaneously determination was rapid, accurate and repeatable.④The elementary results proved that the preparation was stable under the condition of strong irradiance and high humidity, with no significant change in TLC chromatography, contents, as well as aspect. The content of Verbenalin in traditional Verbena officinalis L. yin pian was decreased, thus, flowing Verbena officinalis L. yin pian performed better stability than traditional Verbena officinalis L. yin pian.Conclusion①Optimized technical parameters of flowing Verbena officinalis L. yin pian were all obtained under the condition of pilot scale production. It can instruct industrial production, as well as provide normal standard for the processing procedures.②The flowing Verbena officinalis L. yin pian and traditional Verbena officinalis L. yin pian. showed good equivalence, pill shaped flowing yin pian was good for decoeting.③Quality control method of flowing Verbena officinalis L. yin pian was systematically established. it provides scientific basis for the quality control of flowing Verbena officinalis L. yin pian.④The flowing Verbena officinalis L. yin pian was stable, and convenient for storage.
