Objective To study the protein expression of HGF、C-Met and VEGF in pituitary adenomas, and to evaluate the clinical significance between HGF、C-Met、VEGF and pituitary adenomas. And then discuss the correlation about them.Materials and Methods Tissue samples were obtained from patients who underwent surgery in-Heza Municipal Hospital from February,2008to May,2010. The total number of tissue sample was52, including27males with an average age of40.7±3.1, and25females with an average age of36.3±4.2.All diagnoses of these cases were verified by immunohistochemical pathology. According to the illustration of images and observation in operations, cases were divided into30invasive cases and22noninvasive cases. According to secreting function, cases were divided into32functional cases(PRL16cases, GH5cases, ACTH2cases, multifunctional adenomas9cases) and20nonfunctional cases.The protein expression of HGF, C-Met and VEGF were detected by the S-P immunohistochemical(IHC) method, proceeding according to standard method.5visual fields were chosen randomly to count positive cells under high magnification of200x. The positive staining of HGF, C-Met and VEGF is brown or tan particles located in cytoplasm or cellular membrane. The percentage of positive staining cells in200tumor cells was calculated.The results were evaluated by staining intense and extent in positive cells:negative(-):no positive cells;weak-positive(+):positive cells accounting for less than25%of the total cells; positive (++):positive cells accounting for25-50%of the total cells;strong-positive(+++):positive cells accounting for more than50%of the total cells. Chi-square test was employed to determine the relationship among protein expression of HGF, C-Met and VEGF between the invasive cases and the noninvasive cases. Pearson Rank Correlation was employed to analyze the correlations among protein expression of HGF, C-Met and VEGF in invasive cases.Results The rates of positive expression of HGF in invasive and noninvasive cases were90.0%and40.9%, and there were statistic significance in the two teams respectively (P<0.05). The rates of positive expression of C-Met in invasive and noninvasive cases were86.7%and40.1%, and there were statistic significance in the two teams respectively(P<0.05). The rates of positive expression of VEGF in invasive and noninvasive cases were93.3%and54.5%, and there were statistic significance in the two teams respectively (P<0.05). And the incidence of the invasiveness has no significant difference with the sexuality of patients and secretion of adenomas. There was a positive correlation among protein expression of HGF, C-Met and VEGF in invasive cases, but there was not significant correlation in noninvasive cases.Conclusion The protein expression of HGF, C-Met and VEGF may play important roles in the invasiveness of pituitary adenomas and may be useful makers to predict the invasiveness. The invasiveness growth of adenomas may have connection with high protein expression of HGF, C-Met and VEGF. During invasive growth, there are some correlation among HGF, C-Met and VEGF, reciprocal promotion between HGF and C-Met, HGF and VEGF, C-Met and VEGF. There may have a new research direction of pituitary adenomas’treatment. And the incidence of the invasiveness has no significant difference with the sexuality of patients and secretion of adenomas.
