Objective To detect the expression of MMP-2 ,VEGF and MVD in pituitary adenoma tissues and to investigate the correlation between MMP-2 ,VEGF and MVD and its relationship with the invasiveness of pituitary adenomas. Materials and Methods ⑴Forty-one surgically resected pituitary adenoma samples,including 18 invasive pituitary adenomas and 23 non-invasive pituitary adenomas. All of the tissues were fixed in 10% neutral formalin and embedded in paraffin.⑵Streptavidin-Peroxidase(S-P) immunostaining technique was used to detect MMP-2,VEGF and MVD in all of the tissues. ⑶Relationships between MMP-2,VEGF and MVD were analyzed in pituitary adenoma tissues, and so were relationships between MMP-2,VEGF expression , MVD in invasive pituitary adenomas. Results ⑴The positive expression rate of MMP-2 in invasive pituitary adenomas and non-invasive pituitary adenomas is 72.2%, 26.1% respectively. There is significant difference between 2 groups(p=0.003).⑵The positive expression rate of VEGF in invasive pituitary adenomas and non-invasive pituitary adenomas is 88.9%,30.4% respectively. There is significant difference between 2 groups(p=0.000).⑶There is significant difference of mean value of MVD between invasive pituitary adenomas group(50.51) and non-invasive adenomas group(13.41) (t=8.419,p=0.000). ⑷There is positive correlation between MMP-2,VEGF expression and MVD in invasive pituitary adenomas group(p=0.000). ⑸There is positive correlation between MMP-2 expression and VEGF expression in invasive pituitary adenomas group(p=0.000). Conclusions ⑴Compare with the non-invasive pituitary adenomas , MMP-2 ,VEGF were found high expressed in invasive pituitary adenomas.The invasion of pituitary adenomas is correlate with the expression level of MMP-2 and VEGF.⑵There is positive correlation between MMP-2,VEGF expression and MVD, which suggest MMP-2,VEGF can facilitate pituitary adenomas growth by accelerating angiogenesis.⑶There is positive correlation between the expression of MMP-2 and VEGF. There is a certain correlation between MMP-2 and VEGF.⑷MMPs and VEGF play an important role in invasiveness of pituitary adenomas,The expression of them can be potential invasiveness and prognostic markers of pituitary adenomas.
