Objective To investigate the combination effect of total isoflavones of pueraria(TIP) and vitamin D(VitD) on the osteoporosis(OP) of rat induced by ovariectomizing, in order to elucidate the mechanism and offer the prevention and cure for osteoporosis of post-menopause female.Methods 81 female sprague-dawley (SD) rats of 3-month-old were randomly assinged to sham-operate group and the following 8 experimental groups according to 4×2 factorial design: ovariectomiezd group, ovariectomized but treated with TIP in high, middle and low dose unequally, ovariectomized but treated with VitD and TIP in high, middle and low dose unequally. Rats of the sham-operate group only underwent the resection of a bit fat, but others all underwent the resection of bilateral ovaries. 3 months later, rats of all experimental groups were keep on gavaging with various TIP on and VitD lasting for 3 months. After all rats were killed, we extracted and weight double uteri, calculated the ratio of uteri to body weight of rats. The contents of ALP, BGP, Ca, P, E2 in serum and the length, weight, BMD and Ca content of femurs were examined. the micro-structure of bone were surveyed and analyzed via slice technique and HE staining.Results The deficiency of estrus and descending E2 in serum of SD rats underwent resection of ovaries were detected significantly to sham-operate group(P<0.05), that explained the model of ovariectomized rat significant. The increasing of weight was inhibited in rats of middle, high dose TIP groups, VitD group and low, middle, high dose TIP associating VitD groups(P<0.05), but the interaction of TIP and VitD were not detection. The ratio of uteri to body weight and E2 content in serum increased and indicated synergism between TIP and VitD in rats of middle, high dose TIP groups, VitD group and low, middle, high dose TIP associating VitD groups(P<0.05). The weight and Ca content of femur were increase in rats of middle, high dose TIP groups, VitD group and low, middle, high dose TIP associating VitD groups and so as the length of rats'femur in middle, high dose TIP associating VitD groups(P<0.05). Synergism on the length and Ca content of femur was found in rats conducted with TIP associating VitD. The activity of ALP and BGP decreased in rats of middle, high dose TIP groups, VitD group and low, middle, high dose TIP associating VitD groups and display synergism (P<0.05), the BMD of rats underwent resection of ovaries decreased significantly(P<0.05), while conducted with TIP and VitD the bone density increased(P<0.05).Conclusion1. TIP can improve osteporosis in ovariectomized rat and increase the Ca content and BMD via enhancing the releasing of E2 in serum.2. TIP associating VitD can induce synergism on prevention and cure for osteporosis in ovariectomized rat.
