| AIM Aiming the series of problems were that the processed principle indeterminate, the material of therapeutic effects not clearly, the quality evaluation system is not standard,the lacked of specific processing parameters at improving the stability and control of cuaitan.This paper in the guidance of traditional Chinese medicine theory, using modern science and technology to research the changes of constituents, acute toxicity and pharmacological effects, clarify the principle of cuaitan,and establish the quality standards of cuaitan.METHORDS1.Compare the change rules of chemical composition quantitatively and qualitative between the crude and processed product used with UV,TLC andHPLC analysis.2.Compare the acute toxicity between the crude and the processed product through the experiment of acute toxicity of mice. Research the acute toxicity to rat organs of parts pathological damage degree through the HE staining method.3.The capillary glass tube method was used to compare the effects of blood coagulation of aiye and cuaitan,screen the blood coagulation effective parts of cuaitan. Compared the anti-inflammatory and analgesic effects between the crude and the processed product4.The indicators of processing technology, such as color, properties for external index, the flavonoids contents for inner index, hemostatic effect for pharmacological activity index, were optimized by Lg(34) orthogonal design experiment.A comprehensive evaluation method used to optimization the best processing process of cuaitan, determine the process parameters.5.The contents of flavonoids was activity index, meanwhile, moisture content, extractive of cuaitan under the basis of ten habitats were determined.Then formulate the quality standards of cuaitan.6. Analysis chemical changes before and after processing, combining with the pharmacology experiment preliminary discussion the processing mechanism of cuaitanRESULTS1.Volatile oil, flavonoids, tannic acid,and polysaccharide have different rate reduce after processed,volatile oil and flavonoids chemical composition have significantly changed, TLC map shows ethyl acetate and methanol parts chemical composition parts having great changes.2.The acute toxicity was significantly reduced after processed. Parts of volatile oil having significant effects on rat heart, liver, spleen, lung,and parts of methanol having significant effects on rat liver.3. Pharmacodynamics experiment shows that aiye95%ethanol and flavonoids extracts having significant anticlotting effect compared with blank group;cuaitan water and70%ethanol extracts having significant clotting effect compared with blank group;cuaitan70%ethanol and flavonoids parts having significant clotting effect compared with corresponding parts of aiye. The effective parts of cuaitan having blood coagulation effect are acetone and chloroform extraction parts.The anti-inflammatory and analgesic effects of cuaitan were weaker than aiye,but having significantly effect compared with black group.4. T he optimumly processing method was processed16min at360℃with the amount of sprent vinegar was15%.5. According dry product to calculation, the flavonoids contents was set not less than3.5%, moisture contents was set not more than10%, extracting substances was set not lower than18%.6. The main factors of cuaitan having lower toxicity were contents of volatile oil decreased and chemical composition changed;Cuaitan having better hemostatic effect is the results of anticoagulation function weakened. and coagulation functionint enhancement two aspects interactioned.CONCLUSION After processed, the acute toxicity was significantly reduced,anticlotting effects abated and blood coagulation effect enhanced,which v-erified the the traditional theory of cuaitan having better bleeding effect.This paper has optimumed the best processing craft and formulated the standardization of cuaitan,which could provide the basis for standardized production.This study have significant social benefit and economic benefit. |
