OBJECTIVE:To investigate the association between gene polymorphisms of transcription factor-7like2(TCF7L2) at positions rs7903146and rs12255372and latent autoimmune diabetes in adults (LADA).METHODS:In this case-control study, we chose patients with latent autoimmune diabetes in adults (LADA group, n=47), type2diabetes mellitus(T2DM group, n=100)and normal control subjects (NC group, n=99), and collected clinical characteristics and blood, extracted DNA, PCR amplified target gene, cloned target gene, gene sequencing for rs7903146and rs12255372,and analyzed related genotypes with LADA.RESULTS:(1) In LADA group, T allele frequency and CT/TT genotype frequencies of rs7903146were significantly higher than those in NC group(0.170,0.170,0.085vs0.030,0.040,0.011respectively, P<0.05). Logistic regression analysis showed the CT/TT genotype and T allele were risk factors for latent autoimmune diabetes in adults(OR=2.686,OR=2.462respectively, P<0.001).(2) In T2DM group, T allele frequency and CT/TT genotype frequencies of rs7903146were significantly higher than those in NC group(0.140,0.200,0.040vs0.030,0.040,0.011respectively, P<0.05).Logistic regression analysis showed the CT/TT genotype and T allele were risk factors for type2diabetes mellitus (OR=4.8,OR=4.667respectively, P<0.001).(3) No significant association was observed in rs12255372alleles and genotypes with latent autoimmune diabetes in adults and type2diabetes mellitus.CONCLUSION:T allele polymorphism of rs7903146site in TCF7L2gene might a risk factor for latent autoimmune diabetes in adults (LADA) and type2diabetes mellitus, while polymorphism of rs12255372site in TCF7L2gene may not be a risk factor for latent autoimmune diabetes in adults (LADA) and type2diabetes mellitus.
