Objective:This study aims to investigate the expression level and methylation pattern of IL-17gene in CD4+T cells from patients with latent autoimmune diabetes of adults (LADA) and healthy controls, to primarily research the realationship between the expression level of IL-17and methylation pattern of IL-17gene in CD4+T cells of LADA patients.Methods:PBMCs were isolated from the peripheral venous blood of15patients with LADA and15healthy controls by density gradient centrifugation. CD4+T cells were isolated from the PBMCs using magnetic cell separation technique. The mRNA level of IL-17was measured by real time-PCR. ELISA kit was used to measure IL-17protein concentration in the serum from LADA patients and healthy controls. Bisulfite sequencing was used to determine the methylation status of the IL-17promoter.Results:1.Compared to healthy controls, the expression level of IL-17were significantly higher in patients with LADA(P=0.01);2.ELISA showed serum IL-17level were significantly higher in patients with LADA(P=0.01);3.Bisulfite sequencing results showed the DNA methylation level of the promter of IL-17gene was significantly lower in LADA CD4+T cells than in controls(P=0.02), and it was negative correlated with the IL-17mRNA expression (N=30, r=-0.39, P=0.03).Conclusion:DNA hypomethylation contribute to the overexpression of IL-17in CD4+T cells of LADA patients.
