Objective:To study the effect of an allicin on the expression of MMP-8in the root-canal exudates and in the gingival crevicular fluid with periapical periodontitis and to provide an experimental evidence for the exploitation of allicin as a kind of root canal disinfectant.Method:60teeth involved with periapical periodontitis were collected. They were randomly divided into3groups (n=20).1) Experimental group sealed by allicin in the root canal orifices,2) the control group sealed by calcium hydroxide in the root canal and3) empty control group sealed by saline in the root canal. Collect root canal exudate sampling and the gingival crevicular fluid sampling before and after sealing medicine, detected MMP-8expression levels in root-canal exudates and gingival crevicular fluid by ELISA.Results:1、The results of MMP-8levels in gingival crevicular fluid:The MMP-8concentration D-value in three groups before and after sealing the medication had statistical differences in gingival crevicular fluid. Comparison group:There were significant differences between the experimental group and control group before and after sealing the medication in the root canal of MMP-8levels in gingival crevicular fluid (P<0.05); There was no statistical significance in empty control group (P>0.05).Between the two groups:there were no statistical significance between the experimental group and the control group before and after sealing the medication in the root canal of MMP-8levels in gingival crevicular fluid (P>0.05); there were statistical significances between the experimental group, the control group and the empty control group (P <0.05).2、The results of MMP-8levels in the root-canal exudate:The MMP-8concentration D-value of the three groups before and after sealing the medication had statistic differences in the root-canal exudate.Comparison group:There were statistical significances between the experimental group, the control group and the empty control group before and after sealing the medication in the root canal of MMP-8levels in root canal exudates (P<0.05).Between the two groups:there were no statistical significance between the experimental group and the control group before and after sealing the medication in the root canal of MMP-8levels in gingival crevicular fluid (P>0.05); there were statistical significances between the experimental group, the control group and empty control group (P<0.05).3、The results of MMP-8levels in the root canal exudate and gingival crevicular fluid:Before and after sealing the medication, there were statistical significances between different MMP-8levels in the root canal exudate and in the gingival crevicular fluid of the experimental group and the control group (P<0.05), the MMP-8concentration D-value in the root canal exudates is more than that of the gingival crevicular fluid. There were no statistical significance in the empty control group (P>0.05).Conclusion:1、In periapical periodontitis, the levels of MMP-8will be increased in (?)oth the root canal exudates and the gingival crevicular fluid of the teeth.2、Allicin can decrease the MMP-8levels in the root canal exudates and n the gingival crevicular fluid effectively.3、Both Allicin and Calcium Hydroxide play the same role in reducing he levels of MMP-8.
