Objective:To determine the significance and expression of Caspase-14in cervical carcinoma with different concurrent chemoradiotherapy sensitivities. And to explore the expression of Caspase-14in cervical intraepithelial neoplasis and cervical squamous carcinoma,also with its clinical significance by compared Caspase-3which was essential to apoptosis.Methods:Western blot and immunohistochemistry were used to detect the expression of Caspase-14in cervical carcinoma patients whose sensitivity to Cncurrent Chemoradiotherapy (CCRT) were different. Immunohistochemistry was used to detect the expression of Caspase-14and Caspase-3in273cervical intraepithelial neoplasis and cervical squamous carcinoma.The association of clinical data with Caspase-14and Caspase-3expression was examined.Results:Intensity and rate of Caspase-14expression were higher in the CCRT-highly of sensitive group than in the lowly sensitive group. The results were coincident with Proteomics in preliminary work. The expression of Caspase-3and Caspase-14were both lower in cervical intraepithelial neoplasis and cervical squamous carcinoma than that in normal cervix.The expression of Caspase-14was nearly absent in CIN3and invasive squamous cell carcinoma. Expression of Caspase-3in cervical squamous carcinoma were associated with lymph node(P<0.05), while Caspase-14were not associated with clinical stage,lymph node and patient age (P>0.05). A positive correlation was found between Caspase-and Caspase-14expression in cervical intraepithelial neoplasis and cervical squamous carcinoma (r=0.145, P=0.016).Conclusion:The expression of Caspase-14are associated with concurrent chemoradiotherapy sensitivity in cervical carcinoma and expected to been and candidate biomarker for prediction of sensitivity of concurrent chemoradiotherapy. And the expression of Caspase-14are related with tumourigenesis,but the tumor pathogenesis of Caspase-14in cervical cancer were different with Caspase-3’s. Caspase-14are not associated with the development of cervical carcinoma.
