Objective:To detect the expression of CLU and aspartate-specific cysteine proteases 3,Caspase-3 by immunohistochemistry(Envision)method in normal cervical squamous epithelium tissues,CIN?,CIN??-? and cervical squamous carcinoma tissues,it investigate the relationship between the expression of CLU,caspase-3 and the clinical pathological characteristics of cervical squamous cell carcinoma, and analyze their interaction.Methods Collected 68 cases of samples of cervical lessions that did not receive any radiation or chemotherapy or drug treatment before operation,including 6 cases of normal cervical squamous epithelium tissues 5 cases of CIN?,17 cases of CIN??-? and 40 cases of cervical squamous carcinoma.Immunohistochemistry(IHC)Envision was used to detect the expression of CLU and caspase-3.it explore the relationships between the two proteins' expression and age of patients,clinical stage,tumor size,differentiation degree,vessel invasion,lymph node metastasis and depth of invasion with SCC.All data was disposed and prossesd in SPSS19.0 analysis.Chi-square test or Fisher's exact test were used to identify the difference among groups for frequency data.Wilcoxon singned-rank test was used to identify the difference among groups for Rank data.The relationship was analyze by Spearman rand correlation test. P<0.05 with significance.Results 1.When showed positive,CLU was mainly expressed in the cell cytoplasm or cell membrane of squamous epithelium stained sepia. Caspase-3 was mainly expressed in the nucleus,and some was expressed in the cell cytoplasm,which was stained brownish yellow.2.The positive rate of CLU in normal cervical squamous epithelium?CIN? tissues?CIN??-? tissues and Cervical squamous cell carcinomas tissues were respectively 16.7%(1/6) ? 20.0%(1/5) ? 41.2%(7/17) ? 67.5%(27/40), withescalation. The rate differences of four groups were statistically significant (P<0.05).According to the Spearman rank correlation analysis:the cervical lesion degree and the intensity of CLU expression were positively correlated by rank. Namely, as cervical lesions aggravate, Clusterin expression would enhance(rs=0.987,P=0.013).3.The positive rate of Caspase-3 in normal cervical squamous epithelium?CIN? tissues?CIN??-? tissues and Cervical squamous cell carcinomas tissues were respectively 83.3 %(5/6)?60.0%(3/5)?35.3%(6/17)?25.0%(10/40),with gradual drop. The rate differences of four groups were statistically significant(P<0.05). According to the Spearman rank correlation analysis: the cervical lesion degree and the intensity of Caspase-3 expression were negatively correlated. Namely, as cervical lesions aggravate, Caspase-3 expression would drop(rs=-0.833,P<0.01).4.In cervical squamous cell carcinoma,the expression of CLU was irrelevant with age of patients,clinical stage,tumor size,differentiation degree,vessel invasion,lymph node metastasis and depth of invasion(P>0.05).5.In cervical squamous cell carcinoma,the expression of Caspase-3 was irrelevant with age of patients,clinical stage,tumor size,differentiation degree, vessel invasion,lymph node metastasis and depth of invasion(P>0.05).6.In cervical squamous cell carcinoma,the expression of CLU and Caspase-3 was negatively correlated(rs=-0.762,P=0.001).Conclusions 1.CLU and Caspase-3 might be associated with the genesis and development of cervical squamous cell carcinoma.The expression of CLU would enhance with cervical lesions aggravates and the expression of caspase 3 would drop with cervical lesions aggravates.2.The expression of CLU and caspase-3 is irrelevant with age of patients,clinical stage,tumor size,differentiation degree,vessel invasion,lymph node metastasis and depth of invasion of cervical squamous cell carcinoma. 3.The expression of CLU and Caspase-3 was negatively correlated cervical squamous cell carcinoma.(rs=-0.762,P=0.001).
