The Study On Inflammatory Factors Therapeutic Effect Of DangGuiNianTongTang Pasty With MTX On Adjuvant Arthritis In Rats

Objective: Observing Danggui Niantong Tang pasty’s affection by the adjuvantarthritis (AA), the apoptosis of synoviocytes, serum cytokines and other indicators.Which explore the treatment of rheumatoid arthritis in rats about DangGuiNianTongTangPasty, with the oral administration of methotrexate(MTX), and compared with MTX+Diclofenac Diethylamine Emulgel, to explore the early control of disease progression intopical ointments, and a better understanding of DangGuiNianTongTang Pasty andDiclofenac Diethylamine Emulgel’s difference on relevant indicators and symptoms ofinflammation improve. Methods:(1) Selected wistar rat48, according to the randomnumber table is divided into blank group, model group, treatmentgroup (MTX+DangguiNiantong Decotion of application), and the control group(MTX+Diclofenac DiethylamineEmulgel), n=12, and number, in addition to the blank group, each rat’s right toe skininjected Freund’s complete adjuvant0.1ml proinflammatory.(2)Two weeks after themodeling, all rats by intragastric administration of MTX and gauze wrapped drugs andtopical administration, administration of a total of18days, blank group not be deal with,the model group were given topical poured into distilled water and a simple gauze comforttherapy.(3)Observe the general situation of the rats before and after the modeling,measured by ELISA in IL-17, TNF-ɑ, and take the right side of the knee joint cavitywithin the synovival tissue, and processed into single cell suspension, add the appropriatereagents, and flow cytometry immediately. Results:(1)The treatment group’s TNF-ɑ,IL-17levels lower than the model group (P<0.05), the control group’s TNF-ɑ, IL-17levelslower than the model group (P<0.05), the treatment group’s TNF-ɑ, IL-17level slightlyhigher than the control group (P<0.05).(2)The treatment group’s synovial cell apoptosisrate was higher than the model group (P<0.05), apoptosis of synovial cells in the controlgroup higher than the model group(P<0.05), synovial cell apoptosis rate of the treatment group slightly higher than the control group(P<0.05). Conclusions:(1)The DangGuiNianTongTang Pasty with MTX had good local antinflammatory and analgesic effect, inhibitautoimmune inflammation,restore the abnormal immune regulatory function.(2)TheDangGuiNianTongTang Pasty with MTX had a significant effect in improving the rate ofapotosis of synovial cells.
