Object: To study the genetic polymorphisms of methylenetrahydrofolatereductase(MTHFR) among the Uygur and Han populations in xinjiang;to investigate thepossible association of methylene tetrahydrofolate reductase (MTHFR) genepolymorphisms with adverse effects related to methotrexate (MTX);Methods:(1)radomly collect300healthy people blood sample in People’s Hospital of Xinjiang UygurAutonomous Region;collect31patients blood sample during2010and2011.(2)Thetechnique of polymerase chain reaction and restriction fragment lengthpolymorphism(PCR-RFLP)was usded to detect the genotype and allele frequencies at1298mononucleotide sites of MTHFR among the Uygur and Han populations inXinjiang.(3) Data statistic by the software SPSS17.0statistics. Results:(1)At the site of1298,the C allele frenquencies were found to be12%and25%, for the Uygur and the Hanpopulations respectively, P<0.05, which had statistical differences.(2)The C allelefrequencies at1298mononucleotide sites of MTHFR among the Uygur in Xinjiang hadstatistical differences with those hitherto reported Mao、 Buyi populations inGuizhou.(3)The analysis highlighted a significant association of C677T polymorphismwith MTX toxicity druing Han population.(4)there are no statistically association withoverall MTX toxicity and polymorphism in Uygur patients was detected. Conclusion:(1)The polymorphisms of the1298mononucleotide sites of MTHFR were diverse indifferent populations.(2)The C allele frequencies at the site of MTHFR1298of the Uygurand Han in xinjiang was differneret. The C allele frequencies at1298mononucleotide sitesof MTHFR among the Uygur in xinjiang was different with Mao、Buyi populations inGuizhou because of different nations.(3)These results indicate the association of1298polymorphisms maybe a reduced risk of side effects of MTX in Han Population.
