Objective:Through the test of the brainstem auditory evoked potentials(BAEP), Bcl-2/Bax, and brain-derived neurotrophic factor(BDNF) in Vestibular nuclear area, discussion the Xuanyunding Granules (YXLG) effect of the blood supply insufficiency of Vestibular system in the vascular vertigo rabbit with qi deficiency and blood stasis, in order to Judge the curative effect of the Xuanyunding Granules.Methods:Randomly selecting8rabbits as the blank control group from90healthy adult New Zealand rabbits. The remaining animals with hunger, high fat diet, spin and cervical vertebrae injection modeling. The consistent with the standard model rabbits would be randomly divided into7groups, Xuanyunding Granules groups, Xuanyunding Prescriptions group, Flunarizine group, Xuanyunning group, and model group. Using AABR to test brainstem auditory evoked potentials in rabbits, and immunohistochemical method to detect Bcl-2/Bax, BDNF expression in vestibular nuclear.Result: 1Effect of Xuanyunding Granules on the BAEP in the vertigo rabbits.1.1After the success of modeling the normal group and other groups compared the BAEP. Compared with normal group, each group got obviously prolonged wave or appeared differentiation bad wave. About the V wave and Ⅰ-Ⅴ, the normal group compared with model group, Flunarizine group, Xuanyunning group, Xuanyunding Prescriptions group, Xuanyunding Granules groups, differences were notable significant (P<0.01), suggesting that the vertigo model may affect the supply of blood,and could effect BAEP of the rabbits.1.2Effect of Xuanyunding Granules on the BAEP in the vertigo rabbits. After the drugs intervention,①Compare V wave in each group there were remarkable significant differences (P<0.001), the normal group compared with model group, Flunarizine group, Xuanyunning group there were remarkable significant differences (P<0.001), with Xuanyunding Prescriptions group, Xuanyunding Granules Low group and Middle group there were notable significant differences(P<0.01); with Xuanyunding Granules High group there was significant differences(P<0.05); model group compared with the Xuanyunding Prescriptions group, and Xuanyunding Granules groups differences were notable significant (P<0.01);②each group Ⅰ-Ⅴ wave differences was notable significant (P<0.01), the normal group compared with model group, Xuanyunning group there were notable significant (P<0.001), with Flunarizine group the differences was notable significant (P<0.01), with the Xuanyunding Prescriptions group, and Xuanyunding Granules three groups there were no significance difference (P>0.05), suggesting that Xuanyunding Prescriptions group, Xuanyunding Granules groups could improve the BAEP of the vertigo rabbits, and may affect the supply of blood.2Effect of Xuanyunding Granules on the Bcl-2/Bax of vestibular nuclear in the vertigo rabbits.2.1Effect of Xuanyunding Granules on the Bcl-2of vestibular nuclear in the vertigo rabbits. The average optical density of positive cells in each group have remarkable significant differences (P<0.001), the normal group compared with the other group the average optical density of positive cells were more. Compared with Xuanyunding Prescriptions group, and Xuanyunding Granules groups there was no significant difference (P>0.05), compared with the model group has remarkable significant difference (P<0.001); the model group compared with Xuanyunding Prescriptions group, Xuanyunding Granules High and Middle groups differences were remarkable significant (P<0.001), compared with Xuanyunding Granules Low group differencewas notable significant (P<0.01). The result of Average gray value is opposite. Suggesting the modeling methods can influence the Bcl-2, and the Xuanyunding Prescriptions group, Xuanyunding Granules groups can improve the average optical density of the Bcl-2, and may inhibit the vestibular nuclear area cell apoptosis.2.2Effect of Xuanyunding Granules on the Bax of vestibular nuclear in the vertigo rabbits. Compared with Xuanyunding Prescriptions group, and Xuanyunding Granules groups there were no significant difference (P>0.05), compared with the model group has remarkable significant difference (P<0.001); the model group compared with Xuanyunding Prescriptions group, and Xuanyunding Granules High group differences were remarkable significant (P<0.001), compared with Xuanyunding Granules Middle and Low groups differences were notable significant (P<0.01). The average optical density of Positive cells in each group have remarkable significant differences (P<0.001), normal group compared with Xuanyunding Prescriptions group, Xuanyunding Granules High and Middle groups there were no significant difference(P>0.05), compared with Xuanyunding Granules Low group has significant difference (P<0.05); model group compared with Xuanyunding Prescriptions group, Xuanyunding Granules High and Middle groups there were remarkable significant differences (P<0.001), with Xuanyunding Granules Low group has notable significant difference (P<0.01); Xuanyunding Granules High group compared with Xuanyunding Granules Low group has significant difference (P<0.05). Suggesting the modeling methods can influence the BDNF, and the Xuanyunding Granules groups can improve the average optical density of the BDNF, and may protect neurons in brain.Conclusions:1The method of vertigo modeling with hunger, high fat diet, spin and cervical vertebrae injection would effect of the blood supply in brainstem vestibular system, the brainstem auditory evoked potentials and the factors expression in the Vestibular nuclear area would reflect it.2Xuanyunding Granules can improve the brainstem auditory evoked potentials in the vertigo rabbits, and may improve the blood supply in brainstem vestibular system.3The role of the mechanisms which Yuxianling Granules improve the brainstem auditory evoked potentials and vertigo symptoms in the vertigo rabbits is improved the Bcl-2expression, and reduced the Bax expression, and restraint apoptosis of the neurons.4The role of the mechanisms which Yuxianling Granules improve the brainstem auditory evoked potentials and vertigo symptoms in the vertigo rabbits is improved the BDNF expression.5The effectiveness of Xuanyunding Granules improved the blood supply in brainstem vestibular system is as well as Xuanyunding prescriptions and Flunarizine.
