Objective:To explore the effect of clerodendrum bungei steud flavonoids (CBSF)on the mice with Lewis Lung Cancer, including the general life, the weight of tumor, the tumor inhibition rate and the expression level of p53, bcl-2, bax, to provide the experimental evidence for treatment of Lung Cancer of clerodendrum bungei steud.Method:To set up Lewis lung cancer model with50C57BL/6mice by hypodermic inject, after7days, to choose40successful Lewis lung cancer mice, and randomly divided them into four groups, that model group、DDP group、CBSF high dose group、CBSF low dose group, every groups has10mice, then choose10healthy mice into blank group. Dose method:model group:raperitoneal inject physiological saline0.01ml/g14days continuous; DDP group:raperitoneal inject DDP0.4mg/kg5days continuous; CBSF high dose group:raperitoneal inject CBSF0.6g/kg14days continuous; CBSF low dose group:raperitoneal inject CBSF0.3g/kg14days continuous. Observe the general life of mice in each group. To execute all mice after dose over, weight up the tumors, then calculate the tumor inhibition rate, and use Real-Time PCR method to detect the expression level of p53、bcl-2and bax.Result:1. The effect of general life state:to compare with the blank group, the general life state of each other group has decrease in varied degree, both the general life state of CBSF high dose group and CBSF low dose of group are better than model group and DDP group.2. The effect of the tumor weight(x±s):the tumor weight of four groups as follow:model group is4.70±1.29g, DDP group is2.24±0.68g, CBSF high dose group is3.26±1.06g, CBSF low dose group is3.99±1.31g; the tumor weight of DDP group and CBSF high dose group are lighter than model group(P<0.05or P<0.01),and there is no significant difference between these two groups(P>0.05).3. The tumor inhibition rate (IR):the IR of DDP group is53.4%, the IR of CBSF high dose group is30.6%, the IR of CBSF high dose group is15.1%.4. The effect of gene expression:both CBSF group can up-regulate the expression of p53mRNA and bax mRNA (P<0.05or P<0.01); DDP group can down-regulate the expression of bcl-2mRNA (P<0.05)and up-regulate the expression of bax mRNA (P<0.01).Conclusion:Clerodendrum bungei steud flavonoids can improve the general life state of Lewis lung cancer mice, and inhibit growth of tumor; as well as that the antineoplastic mechanisms may be to induce tumor cell apoptosis by up-regulate the expression of p53mRNA and bax mRNA.
