Experimental Study Of The Four Traditional Chinese Medicine On The Proliferation Of Rat Bone Marrow Mesenchymal Stem Cells Into Chondrocytes

OBJECTIVE:The observation of four Chinese medicine (Astragalus, Epimedium, snow lotus and Yougui drink) on the proliferation of rat bone marrow mesenchymal stem cells (BMSCs) to chondrogenic differentiation.METHODS:This study is divided rat bMSCs separation, culture, subculture and proliferation; induced to differentiate into cartilage-like cells and proliferation by transforming growth factor-β1(TGF-β1) in vitro observation of Astragalus, Epimedium, snow lotus and Yougui drink onby the proliferation of cartilage-like cells after induction of TGF-β1.RESULTS:Adherent method successfully isolated and cultured get rats bMSCs, cell morphology is a good, vigorous growth, cartilage-like cells the rat bMSCs induced by conditioned medium, immunohistochemical staining to observe the expression of collagen type Ⅱ, Astragalus, Epimedium, snow After the Lotus and Youguiyin the intervention, MTT assay were measured and statistical analysis showed that Astragalus Decoction can obviously promote cell proliferation, Youguiyin decoction of the four concentration groups have shown that inhibition of cell growth, Epimedium fried both agents to promote have inhibitory effect displayed worthless, the Saussurea alcohol extracts only10"5concentrations promote cell proliferation. The four Chinese medicines containing serum studies have shown that astragalus drug serum on cell proliferation in15%and1%concentrations of the of Epimedium drug serum concentration of15%and5%group display, the snow lotus drug serum10%,5%and1%three concentrations promote cell proliferation, Yougui drink the serum of four concentrations of drugs have shown to promote cell proliferation.CONCLUSION:Four kinds of traditional Chinese medicine to promote chondrogenic differentiation, the proliferation of only10-5snow lotus alcohol extraction liquid group, but also indicate that the four Chinese medicines containing serum has a role in promoting the proliferation of MSCs into cartilage differentiation.SIGNIFICANCE:Retrieve relevant documents, Yougui drink and the Astragalus drug decoction and snow lotus alcohol extracts to observe the cartilage-like cell proliferation induced by TGF-β1has not been reported by experimental studies, suggesting that four Chinese medicines containing serum on MSCschondrogenic differentiation, proliferation role in promoting new ideas into chondrocytes in the cartilage defect treatment.
