The Effects Of Compound Traditional Chinese Medicine Of Ginseng And Astragalus With5-FU Inhibited Proliferation And Induced Apoptosis In Human Gastric Carcinoma MGC-803Cells

Objective To observe the effect of compound traditional Chinese medicine (TCM) of Ginseng and Astragalus with5-FU alone and combination are on the role of cell growth inhibition, cell colony formation, induction of apoptosis and cell cycle in gastric cancer MGC-803cells in human.Methods The cell proliferation inhibition was detected by MTT assay, the median effective concentration of different drugs used alone and combination were analyzed by median effect equation, the cell cycle and apoptosis was observed by flow cytometry, the cell colony of formation was detected by Giemsa staining.Results The inhibition rate of cancer MGC-803cells increase when the drug concentration of compound TCM Ginseng and Astragalus with5-FU were used alone and their combination increasingly. The median effective concentration of the two drugs combination was less than the combination of the median effective concentration of different drugs alone were analyzed by the median effect equation. The differences of inhibitory rates of combination group, traditional Chinese medicine group and5-FU group used alone were statistically significant(P<0.05). The median effective concentration of combination group on the role of promote apoptosis, inhibition of cell colony formation, blocking cells in G0/G1phase are significant. The difference of apoptosis rate, inhibition of cell colony formation rate,G0/G1phase in combination group, traditional Chinese medicine group and5-FU group used alone are statistically significant(P<0.05).Conclusion Compound TCM of Ginseng and Astragalus,5-FU combination is better than the two drugs alone on the role of inhibition of cell growth and cell colony formation, induction of apoptosis and blocking of the cell cycle at G0/G1stage in gastric cancer MGC-803cells in human.
