Objective:Cervical cancer is one of the most common malignant tumors, About130,000new cases a year in our country,ranks the second in the gynecological malignant tumor after breast cancer.cervical cancer has a series of precancerous lesions, Atypical hyperplasia and carcinoma in situ are typical forms of cervical precancerous lesions.it also be called cervical intraepithelial neplasia (CIN). cervical intraepithelial neplasia is divided into three levels,that is CIN Ⅰ, CIN Ⅱ,CINⅢ,the occurrence,development and formation of cervical cancer is a quantitative change to qualitative change process.Any degree of cervical precancerous lesions may develop into invasive carcinoma of the cervix. Classified by Cytopathology, cervical cancer cells can be divided into:high differentiation, differentiation, low differentiation, the cell differentiation degree is closely related to the prognosis of cervical cancerAlthough,cervical cancer is the woman’s common malignant tumor in our country,deprived ten thousands of women’s lives, and patient’s age tended to be younger. However,so far there is still a lack of early diagnosis of cervical cancer and prognostic indicators Of idealTPX2is a candidate cancer gene discovered recently.regulate by thecell cycle strictly,and play an important role in cancer cell’s increment.Therefore TPX2protein may play an important role in the evolution of development.However,the expression of TPX2in cervical cancer have not been reported.The purpose of my study:1Objective To study the expression of TPX2protein and mRNA in human cervical squamous carcinoma, and the relationship between its expression and clinicopathologyical features of cervical cancer. 2To search for the new indicators of diagnostic of cervical cancer.Methods:1from October2009to November2010in the Fourth Hospital of Hebei Medical University,Immunohistochemistry、reverse transcription polymerase chain reaction were used to analyze the expression levels of TPX2mRNA and protein respectively, in fresh invasive cervical squamous cancer (n=62)、 normal cervical uteri (n=15) and CIN tissues(n=30).The specimens are isolated within half an hour in vitro.kept in liquid nitrogen and then save in the refrigerator-80℃2Expression of TPX2mRNA was detected by RT-PC in62cases of cervical cancer tissue and15cases of normal cervical tissue.3Statistical analysis. SPSS13.0statistical software was used for statistical analyses. Statistical data are presented as mean±standard deviation. Student test was used to compare two samples, Kruskal-Wallis test for group comparisons, as well as the Mann-Whitney U test for nonparametric independent two-group comparisons were performed. A P-value of<0.05was considered as statistically significant.Result:1Detection of TPX2protein in different tissues of the cervix by immunohistochemistry. The expression of TPX2protein was localized in the cell nucleus. The expression of TPX2protein in CIN and cervical carcinoma tissues gradually increased, however, no expression in normal cervix tissue was found. There were significant differences among the three groups (P<0.01)(Fig2and Table1).2The expression of TPX2:By the rank sum test, the expression intensity of TPX2have no significant difference between the group of the age≤40years old and the group of the age>40years old.(P>0.05).In clinical stages: the expression intensity of TPX2have a remarkable difference between the group stage Ⅰ and Ⅱ(P<0.05).In pathology graduation:the expression intensity of TPX2have a remarkable difference between the high differentiation group and the middle and high differentiation group. Lymph node metastasis:the expression of TPX2in the tissue which lymph node metastasised,and have no relationship with the age of patients.3By RT-PCR, there was no expression of TPX2mRNA in normal cervical tissues, The expression of TPX2mRNA cervical carcinoma is0.441±0.011, respectively. There was significant difference between the two tissues (t=-19.568,P<0.01).Conclusion:1The expression of TPX2have no correlation with the age of patient.It is prompted that TPX2is not a factor for cervical cancer younger.2The expression of TPX2have no relationship with the FIGO stage,pathological grade,lymph node metastasis.TPX2may play an important role in the development,invasion,metastasis of cervical cancer.
