Objective: To study the expression of TPX2 and Aurora-B proteins in different human cervical tissues(normal cervical tissues,intraepithelial neoplasia,cervical squamous cell carcinoma),and to analyze the relationship between the expression of TPX2 and Aurora-B proteins in cervical squamous cell carcinoma and the clinicopathological parameters of patients and their correlation.The relationship between the expression level of both and the radiotherapy sensitivity of cervical squamous cell carcinoma was investigated.Methods: 1.According to the inclusion criteria of this study,60 patients with stage IIB-IVA cervical squamous cell carcinoma diagnosed pathologically in our hospital from October 2017 to September 2019(both newly treated patients,aged 30-74 years)were selected as experiments.Group,normal cervical tissue and 30 cases of cervical intraepithelial neoplasia(CIN)tissue were used as a control group.The expressions of TPX2 and Aurora-B protein in different cervical tissues were detected by immunohistochemistry,and TPX2 was analyzed in cervical squamous cell carcinoma tissues.The relationship between the expression of Aurora-B protein and patient's age,clinical stage,degree of pathological differentiation,lymph node metastasis and their correlation.2.Simultaneous chemoradiotherapy was performed on the above 60 patients with cervical squamous cell carcinoma.The radiotherapy method was external pelvic irradiation combined with intracavitary irradiation.The chemotherapy was platinum monotherapy.All patients received magnetic resonance imaging before and 1 month after radiotherapy Gynecological examination,according to the WHO solid tumor efficacy criteria to evaluate the short-term efficacy,divided into high-sensitivity group(CR)and low-sensitivity group(PR + NC),according to the above group,the results of TPX2 and Aurora-B protein in squamous cell carcinoma specimens They were also divided into two groups,and their relationship with radiosensitivity was analyzed.Results: 1.The positive expression rates of TPX2 protein in normal cervical tissue,CIN and cervical squamous cell carcinoma tissues were 6.67%,76.67%,and 86.67%,respectively.Thepositive expression rates of Aurora-B protein in these three groups of cervical tissues were6.67%,50.00%,73.33%,the difference between the three groups was statistically significant(P <0.05).Further pairwise comparisons revealed that the expressions of TPX2 and Aurora-B proteins in cervical squamous cell carcinoma and CIN tissues were significantly higher than those in normal cervical tissues(P <0.05).There was no significant difference between cervical squamous cell carcinoma group and CIN group(P > 0.05).2.The expression level of TPX2 protein in patients with cervical squamous cell carcinoma of stage IIIB-IVA of FIGO is higher than that of stage IIB-?A(P <0.05),and the expression intensity of the medium-low differentiation group is higher than that of the highly differentiated group(P <0.05).The expression intensity of lymph node metastasis group was higher than that of no lymph node metastasis group(P <0.05).The expression intensity of Aurora-B protein in cervical squamous cell carcinoma patients in the low-differentiated group was higher than that in the high-differentiated group(P <0.05),and there was no significant difference with FIGO stage and lymph node metastasis(P> 0.05);There was no significant correlation with age(P> 0.05).Correlation analysis showed that the expressions of TPX2 and Aurora-B proteins were positively correlated in cervical squamous cell carcinoma tissues(r = 0.456,P <0.05).3.The expression intensity of TPX2 and Aurora-B proteins in the low-sensitivity group was higher than that in the high-sensitivity group(P <0.05).Conclusions: 1.TPX2 and Aurora-B protein are highly expressed in cervical squamous cell carcinoma.The expression of TPX2 protein is related to the clinical stage of cervical squamous cell carcinoma,the degree of tissue cell differentiation and lymph node metastasis.Aurora-B protein is related to the degree of pathological differentiation.And the expression of the two is positively correlated,and its positive expression can be used as an important indicator for the development of cervical squamous cell carcinoma and evaluation of the disease.2.TPX2 and Aurora-B proteins are related to the radiosensitivity of cervical squamous cell carcinoma,and their overexpression will cause radiotherapy resistance,which is of great significance in formulating personalized treatment for patients.
