Purpose: This is a study on chemical constituents from the leaves of Cyclocaryapaliurus (Batal.) Ijinakaja. The key point is to find and investigate anti diabetic parts andconstituents isolated from effective parts of Cyclocarya paliurus. On the basis of structuralelucidation, the anti diabetic activity of these compounds will be evaluated.Method: By the extraction and separation techniques of water extraction, organicsolvents dissolution and macroporous resin column chromatography and inhibitionexperiments in vitro of protein nonenzymatic glycosylation and α glucosidase, theanti diabetic parts of the leaves of Cyclocarya paliurus were screened out. Applying thechromatographic methods including silica gel, ODS, sephadex LH20and prep HPLC, etc,compounds were isolated from the active parts. Their structures were elucidated byphysic chemical methods and1D NMR,2D NMR and ESI MS techniques and theiranti diabetic activities were evaluated.Result: Four extractions and compounds II、III、IV、V、VII of Cyclocarya paliurus couldinhibit protein nonenzymatic glycosylation in various degree. Meanwhile, α glucosidaseactivity could be obviously inhibited on30%ethanol extracted by macroporous resin columnchromatography. Seven compounds were isolated from the chloroform/methanol dissolvingpart of Cyclocarya paliurus. Except for unidentified compound VI, compound I waselucidated as a mixture of arjunglucoside II (I A) and quadranoside IV (I B) and the other fivecompounds were identified as cyclocarioside I (II);(20S,24R) epoxydammarane (3β,12β)25hydroxyl12O β D quinovopyranosyl3O (5′O acetyl) α L arabinofuranoside (III);(20S,24R) epoxydammarane (3β,12β)25hydroxyl12O β D quinovopyranosyl3O α L arabinopyranoside (IV);(20S,24R) epoxydammarane (3β,12β)25hydroxyl12O α L arabinopyranoyl3O α L ara binofuranoside (V);(20S,24R) epoxydammarane (3β,12β)25hydroxyl12O β D glucopyra noyl3O α L arabinofuranoside (VII).Conclusion: The anti diabetic parts of the leaves of Cyclocarya paliurus could betriterpenoid saponins. Compounds III, IV, V and VII are new compounds.
