Effects Of Cyclocarya Paliurus (Batal.) Lljinskaja Polysaccharide On Antioxidant Activity And Lipid Metabolism-related Genes In Rats

This paper was conducted to study the effects of Cyclocarya paliurus(Batal.) Iljinskaja polysaccharides(CPP) on lipids metabolism and PPARγ, PPARδ, CAP, C/EBP genes expression in hyperlipidemic Rats. The hyperlipidemic rats were randomly divided into model control, CPP high dose,middle dose, low dose groups, simvastatin, CPP water group(16 rats per group). After 8 weeks of treatment, the antioxidant effect of Cyclocarya paliurus(Batal.) Iljinskaja polysaccharide(CPP) in high-fat emulsion-induced hyperlipidaemic rats was detected. Also, the antioxidant enzymes and the activity of lipid metabolic enzyme in rats liver were elevated. The expressions of PPARγ, PPARδ, CAP,C/EBP were investigated by Q-PCR to explore the effect of Cyclocarya paliurus(Batal.) Iljinskaja polysaccharides in liver and white adipose tissue of hyperlipidemic rats. The protein expression of related genes were performed by westen-blotting method. The results showed as follows:(1) Cyclocarya paliurus(Batal.) Iljinskaja polysaccharides significantly increased the total antioxidant capacity(T-AOC), catalase(CAT), glutathione peroxidase(GSH-Px) and total superoxide dismutase(T-SOD) activities with the highest rate of 73.49%, 48.63%, 10.08% and 17.64%,respectively, while decreased the malondiadehyde(MDA) level with the decreasing rate of 49.61% in the liver;(2) Cyclocarya paliurus(Batal.) Iljinskaja polysaccharides significantly restrained the fatty acid synthase(FAS), acetyl-CoA carboxylase(ACC) and peroxisomes proliferator-activated receptor-γ(PPARγ) activities with the highest rate of 44.57%, 58.56% and 50.63%, respectively, while increased the hormone-sensitive lipase(HSL), liportein lipase(LPL), adipose triglyceride lipase(ATGL) and leptin(LEP) level with the increasing rate of 74.45%, 45.32%, 43.92%, 89.47% in the liver;(3) Cyclocarya paliurus(Batal.) Iljinskaja polysaccharides decreased the peroxisomes proliferator-activated receptor-γ(PPARγ), CCAAT/enhancer binding proteins(C/EBP) mRNA expression levels(P<0.05 or P<0.01), in the liver the highest growing rate was 46.78%, 58.44% and in the adipose the highest growing rate was 41.25, 55.88%, respectively. In addition, Cyclocarya Paliurus(Batal.) Iljinskaja polysaccharides could improve the proliferator-activated receptor-δ(PPARδ) and Cbl-associated protein(CAP) mRNA expressions in the liver and adipose with the falling rate of151.04%, 272.5% and 59.99%, 660.78%;(4) Cyclocarya paliurus(Batal.) Iljinskaja polysaccharides could not significantly affect the protein expression of peroxisomes proliferator-activated receptor-γ(PPARγ), peroxisomes proliferator-activated receptor-δ(PPARδ), Cbl-associated protein(CAP) and CCAAT/enhancer binding proteins(C/EBP).Our results indicated that CPP has inhibitory effects on the lipid oxidation in the liver, regulates the gene expression related to the lipid metabolism, thus to ease the hyperlipidemia.
