Objective: Zebrafish diabetic model was established,and used to evaluate the hypoglycemic efficacy of cyclocarya paliurus(Batal.)Iljinskajain.Methods: Zebrafish diabetic model was established by experiment to explore the combination concentration of alloxan and sucrose,and using the four clinical commonly used medications: metformin,glibenclamide,glimepiride,and acarbose for model validation.The cyclocarya paliurus produced from Hubei Suizhou city and Wenzhou Wencheng County were used to prepare the solution with hot water immersion method for evaluation of hypoglycemic effect.(1)Evaluation of hypoglycemic effect of cyclocarya paliurus: on 4 day post fertilization(4dpf),AB wild type zebrafishes were treated by a combination of 1 mM alloxan and 4%sucrose to establish diabetic zebrafish model,and the fishes were followedly treated with0.50,1.00 and 2.00 mg/mL cyclocarya paliurus solution.Positive control group were administrated with 0.01 mg/mL metformin,and a normal control group and a diabetic model control group were set up.After the treatment,glucose levels of zebrafishes in each group were measured with a blood glucose meter.(2)Evaluation of effect of cyclocarya paliurus on ? cell regeneration: after an establishment of 4 dpf ? cell fluorescence diabetic zebrafish model by a combined treatment of 1 mM alloxan and 4% sucrose,the zebrafishes were treated with 0.50,1.00 and 2.00 mg/mL of cyclocarya paliurus solution.Positive control group were given with 0.01 mg/mL metformin,and a normal control group and a diabetic model control group were set up.At the end of treatment,the fluorescent intensity of zebrafish ? cells was measured to analyze the regeneration of ? cells.Results: 4 day post fertilization(4 dpf),AB wild type zebrafishes were treated by a combination of 1 mM alloxan and 4% sucrose for 24 h success to establish diabetic zebrafish model.Metformin,glibenclamide,glimepiride,and acarbose had an obvious hypoglycemic effect in zebrafish diabetic model,this results were consistent with the clinical practice.Cyclocarya paliurus produced from Hubei Suizhou city and Wenzhou Wencheng County had obvious hypoglycemic effect and promoted the regeneration of ?cells in a dose-dependent manner.Cyclocarya paliurus from Hubei Suizhou city played a better role than that of Wenzhou Wencheng County in the hypoglycemic effect and ? cell regeneration at high doses.The results suggest that cyclocarya paliurus plays a hypoglycemic role and it showes a difference of geographic region.
