Background: Cervical caner is the most common cancer in all femme malignant tumors,whose morbidity increased gradually by years, the younger patient ratio is more. Thereare about130,000new patients in China every year, and It is seriously threatening thewomen’s health. The high-speed growth of the solid tumor will cause relativeinsufficiency of internal blood supply and decreased oxygen tension. how judge whetherthe tumor tissue existence lacks oxygen becomes the hot spot that the cancer studiescurrently. Hypoxia inducible factor-1alpha (HIF-1α) is a hypoxia-responsive factor,which commonly exists in the tissues of solid tumors. It plays crucial roles inmaintaining cell energy metabolism, tumor angiogenesis and metastasis, cellproliferation and apoptosis.。carbonic anhydrase IX(CAIX)is a new-found carbonicanhydrase household(CAs)different constructions,which is a kind of new tumor antigenand have a latent function in the middle of regulating cell enzyme amplification andtumor formation, and contain closely related metabolism with hypoxia of tumor. theresearch of and HIF-1α and CAIX organizes a series of variety and evaluation that thetiny environmenthypoxia causes to undoubtedly have important meaning in thediagnosis of the cervical cancer medical treatment to the cervical cancer. Currently therelativity research between HIF-1α and CAⅨ is less, especially the relativity still haveno related report in the expression in the cervical cancer in later period. Objective: Toinvestigate the expressions of HIF-1α and CAIX in normal cervix tissues, cervicalintraepithelial neoplasia (CIN) and cervical squamous cell carcinomas (CSCC) ofdifferent stages,we observe their expressions in different cervix lesions, to investigatethe change trend from normal cervix tissue-precancer lesion-cancer process, and toinvestigate the relationship between HIF-1α and CAIX in cervical squamous cellcarcinomas. Method: Immunohistochemical analysis was used to detect the expressionof HIF-1α and CAIX in normal cervix tissue,CINI,CINII,CINIII and cervical squamouscell carcinoma of10cases,15,16,17,35cases each, which had been histologicallyconfined and were divided into control group and experimental group. the correlation ofthem with the histological grade, and differentiation stages were analyzed statistically, atthe same time the relationship between HIF-1α and CAIX was studied in thedevelopment of cervical squamous cell carcinoma for a preliminary study. Result: 1.The positive expression rate of HIF-1α in normal cervical tissue, CIN, cervicalsquamous cell carcinoma were10.0%,57.5%,77.1%,the trend has graduallyincreased(P<0.05). Among them,significant difference was existed between normalcervical group and CIN group(P<0.05),also between normal cervical group and cancergroup(P<0.05), The positive expression rates of HIF-1αwere51.5%,66.7%,91.3%inCINI,CINII,CINIII, respectively. There was significantly difference between thedifferent groups(p<0.05). HIF-1α expression was significantly higher in CINII andCINIII than in CINI(p<0.05).2.The positive expression rate of CAIX in normal cervicaltissue, CIN, cervical Squamous cell carcinoma were6.67%,54.05%,83.33%,the trendhas gradually increased(P<0.05). Among them,significant difference was existedbetween normal cervical group and CIN group(P<0.05),also between normal cervicalgroup and cancer group(P<0.05),The positive expression rates of CAIX were51.5%,66.7%,91.3%in CINI,CINII,CINIII, respectively. There was significantly differencebetween the different groups(p<0.05). HIF-1α expression was significantly higher inCINII and CINIII than in CINI (p<0.05).3. The expressions of HIF-1α and CAIXwere significantly correlated with clinical stage of cervical squamous cell carcinoma(X2=7.29,P<0.01);differentiation degree of tumor cells(X2=4.61,P<0.05);Theexpression of CAIX was significantly correlated with clinical stage of cervicalsquamous cell carcinoma(X2=7.29,P<0.01);differentiation degree of tumor cells(X2=4.61,P<0.05).4.There was a positive correlation between CAIX and HIF-1α(r=0.69, P<0.05).Conclusion: Conclusion:HIF-1αand CAIX may play an significantroles in occurrence and development of cervical squamous cell carcinoma, both of themmay play synergetic roles. So combined detection of expressions of HIF-1α and CAIXcan be regarded as an important index to judge malignancy of the tumor and evaluateprognosis of cervical squamous cell carcinoma.
