Time-resolved fluoroimmunoassay (TRFIA) is a novel supermicro analysis in vitro usinglanthanide chelate as detectable label. In recent years, TRFIA have been widely used inclinical diagnosis and analyses, life science, and medical science because of its much highersensitivity and accuracy than other radioimmunoassay methods. It has become a verypromising analytical tool.The synthesis of bifunctional chelator is the key of timed-resolved fluorescenceimmunoassay. The chelator should not only conjugate with proteins by covalent bonds, butalso chelate with rare-earth ion. The purity and the chelating property of bifunctional chelatoris the key to the precision and sensitivity of the measurements.In this dissertation, there was a big paragraph discussed the synthesis ofN~2-(p-isothiocyanato benzyl)-diethylenetriamine-N~1, N~1, N~3, N~3-tetraacetic acid, which belongto N~2-(p-isothiocyanato benzyl)-DTTAEuNa. The condition of the synthesis was researched.We studied the temperature time, catalyst amount, ratio of adding material, and proportion ofmaterial of the reactions. As a result, the method of the synthesis was improved, and the yieldwas enhanced.
