Objective: The HPLC fingerprint and the ISSR-DNA fingerprint of Maohuacultiva were established to analyze the correlation of its common peaks, chemicalconstituents and genetic diversity. Thus providing theoretical evidence and data forquality assessment,identification and for screening of good germplasms.Methods: The HPLC fingerprint of Maohua cultiva was studied. And fourActive constituents were simultaneously analyzed by the HPLC method. ISSRtechnique was used to establish the ISSR-DNA fingerprint. Study the correlationbetween HPLC fingerprint, the content of active constituents and ISSR-DNA bandscharacters of Maohua cultiva by Similarity evaluation and cluster analysis.Results: Based on chlorogenic acid, the HPLC fingerprint of Maohua cultivawas established, there were13common peaks. Differences were existed in the HPLCfingerprint of different germplasm rescources of Maohua cultiva.Active constituentsamong the ten germplasms were existed.15primers screened out were used for ISSRamplification.A total of307bands were obtained including217polymorphic loci. Theaverage polymorphic frequency was70.68%. The genetic similarity coefficientsranged from0.6456-0.8987among the germplasms. Dendrogram revealed that tenMaohua cultiva were divided into two classes: the first category was only consisted ofXM-MH-1, other samples were clustered into the second class, second class wasdivided into three groups.Conclusions: There are certain correlations between the genetic variation andchemical constituents, thus providing a theoretical foundation for quality assessmentand for the identification and classification of honeysuckle cultivars.
