Objective:The study is to establish a bioactivity assays of luteinizing hormone, and the bioactivity of LH in serum samples be detected by LH bioactivity assays.Methods:Human or SD rats Serum samples be used to stimulate rat Leydig cells and produce testosterone in vitro culture, and LH levels were measured by testosterone levels which were detected by ECLI.The bioactivity of rats and human serum LH were detected by LH bioactivity assays. Twenty-four of two-month-old male SD rats were randomly divided into three groups:Quiet control group, moderate-intensity exercise group (28m/min) and high-intensity exercise group (35m/min). The exercise group exercise respectively for eight weeks, six times for a week, and40minutes at every turn in the treadmill. The blood samples were collected after8weeks. Human serum samples were collected from21healthy men who are selected randomly. Healthy men are the two different grounds, young people and older people. SD rats and human serum samples be used to stimulate rat Leydig cells and produce testosterone which could measure bioactivity of the serum LH.Results:1)After4h incubation of Leydig cells, the testosterone concentration was at the maximum. It shows that it is the optimum time for four hours.2) It is the parallelism with the dose-response curves for serial dilutions of the human serum and SD rats serum. These suggest that Leydig cells have a excitant reaction by exogenous LH. Bioactivity assays of LH was used to detect bioactivity of serum samples.3) Compared with the quiet control group, bioactivity of serum LH declined in the ME group and the HE group (p<0.01). It wasn’t different significantly about bioactivity of serum LH of the ME group and the HE group (p>0.05).4) Compared with the young people, bioactivity of serum LH declined in the elderly people (p<0.01) Conclusions:The bioactivity assays of LH be established. Bioactivity of the exercise rats serum LH declined. Bioactivity of serum LH in the elderly people is lower than the young people.
