ObjectiveIschemic cerebrovascular disease is a common and frequently occurring disease in elderly,which has high incidence morbidity and death rate. With the developing of aging population,the hazard to human health caused by ischemic cerebrovascular disease have become more prominent. Then,how to improve the treatment of ischemic cerebrovascular disease has become one of the most important topics in the world medical profession nowadays.With the indepth study and better understanding of ischemic cerebrovascular disease,scholars find out that Ischemia and hypoxia stimulation will triger the angiogenesis mechanism and promote the formation of ischemic penumbra’s collateral circulation.However,the compensateory blood flow is not sufficient to restore normal blood supply,as the ischemia and reperfusion injury followed by.TAG(therapeuticangiogenesis),a treatment that using of artificial means not only to induce the angiogenesis of ischemic surrounding tissue and the formation of collateral vessel,but also to improve tissue ischemia,opened up a new approach for the treatment of ischemic cerebrovascular disease.The process of angiogenesis is ragulated by a series of both angiogenesis-promoting factor and angiogenesis inhibitory factor.Either the bFGF(basic fibroblast growth factor) effected as angiogenesis-promoting factor or the ES(endostatin) effected as angiogenesis inhibitor plays an important role in the the process of angiogenesis.In this subject,we made MCAO model by using modified suture method.We observed the effection of electroacupuncture on bFGF(basic fibroblast growth factor),ES(endostatin) and CD34’s expression of focal cerebral ischemia and reperfusion of ischemic rat brain tissue.We discuss the possible mechanism trigger by electro-acupuncture effcted on cerebral blood reperfusion in rat angiogenesis from the perspective of angiogenesis. We aimed on providing the experimental evidence for more effective clinical treatment of ischemic cerebralvascular diseases.Methods80SD rats were randomly divided into four groups:Normal group, shame operation group,model group (ischemia1h reperfusion12h,24h,48h),el-ectro-acupuncture group(1h ischemia and reperfusion12h,24h,48h,plus EA tr-eatment),10rats in each group.We use modified Longa Prepara the focal cere-bral ischemia and reperfusion model (MCAO).Reference to Zea-Longa5gra-des standard,we rate and screen the nerve dysfunction in experimental animals. We only retain the neurological dysfunction in rats upper than grade one.EA g-roup stimulated in rats’"GV20","GV26",bilateral"ST36"by disperse-dense waves electro-acupuncture.EA group rats’first stimulated time was2h after cerebral ischemia and reperfusion,as other goups were12h after,then each group stimulated once after every24h and sitmulater the last time1h before cardiac perfusion fixation.We detected the ischemic rats’brain tissue of bFGF,ES positive expression level and microvessel density by SABC(Strept Avidin-Biotin Complex). We used the sofware SPSS17.0in data processing.All statistical results are expressed as Mean±standard deviation (x±s), significant criteria P<0.05. Results (1)Normal and sham operation:bFGF,ES,CD34showed in the brain vascular endothelial cells were low expression or no expression.Showed no significant difference between2groups(P>0.05).(2)The model groups’bFGF,ES,CD34expression was significantly increased in each period.Compared with the normal group and sham group,there are significant differences(P<0.05).(3)Each model group and EA group of bFGF,ES expression and CD34expression of microvascular were related.(4)Comparing the corresponding phase group between ischemia electro-acupcupuncture group and model group,bFGF expression was significantly enhanced,were significant differences(P<0.05).ES expression was significantly diminished,were significant differences(P<0.05). CD34expression enhanced, were significant difference. Conclusion(1)There is no significant effect made by general surgical trauma on the brain tissue bFGF expression of the ES and the number of microvascular density.(2)Cerebral ischemia and hypoxia promote the increase the expression of bFGF, ES,and microvascular density in brain tissue.(3)EA can promote a significant increase the expression of bFGF around cerebral ischemia and reperfusion in the rat brain infarct.And significantly decreased ES expression.(4) EA could promote the number increasing of microvascular cerebral ischemia and reperfusion in the rat brain.
